貉细小病毒RDPV-JL3株的分离鉴定及VP2基因序列分析

摘要 本试验从吉林省某养殖场疑似患貉细小病毒的貉肠道组织中成功分离出1株貉细小病毒(Raccoon dog parvovirus,RDPV),将其命名为RDPV-JL3株。对其VP2基因进行克隆测序和序列分析,结果表明,VP2核苷酸基因序列与RDPV HeB10-3(KJ194463)株相似性为100%,属于CPV-2亚型。系统进化树表明其与RDPV HeB10-3株和RDPV-17-JL-9(MH581183)株处于同一进化分支且与CPV-Vac1疫苗株亲缘关系密切。本研究为更好地在本地区开展貉细小病毒流行病学调查提供了有益借鉴,也为深入研究貉细小病毒变异和传播的分子机制奠定了基础。 A strain of raccoon dog parvovirus(Raccoon dog parvovirus,RDPV)was successfully isolated from the suspected raccoon dog parvovirus infection in Jilin province,it was named the RDPV-JL3 strain.The VP2 gene of the isolate was cloned,followed by sequence analysis.Results showed that the nucleotide sequence of VP2 isolate was 100%similar to that of RDPV HeB10-3(KJ194463.1).Key amino acid sites analysis revealed that the isolate belonged to the CPV-2 type.The phylogenetic annlysis showed that the RDPV-JL3 strain was clustered with RDPV HeB10-3(KJ194463.1)and RDPV-JL-17-9(MH581183.1).Meanwhile,the isolate’s genetic relationship was found it was close to that of CPV-Vac1,as evidenced by the phylogenetic tree.This study not only provides a useful reference for better epidemiological investigation of regional raccoon dog parvovirus,but also facilitate the further studies of the molecular mechanisms by which the raccoon dog parvovirus mutates and/or transmits.
