摘要
为研究葛根护肝片对慢性酒精性肝损伤大鼠的保护作用,采用酒精灌胃量阶段性递增的方式4周制备慢性酒精性肝损伤大鼠模型,随机分为模型组、阳性药组(葛根枳椇子胶囊333.3 mg·kg^(-1))及葛根护肝片高(520.8 mg·kg^(-1))、中(260.4 mg·kg^(-1))和低(130.2 mg·kg^(-1))剂量组,另设正常对照组,连续给予各自受试物4周。末次给予受试物后,测定血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)的活力总胆红素(TBIL)、总胆固醇(TC)、甘油三酯(TG)及低密度脂蛋白胆固醇(LDL-C)的含量,谷氨酸脱氢酶(GLDH)、α-谷胱甘肽-S转移酶(α-GST)、嘌呤核苷酸磷酸化酶(PNP)和精氨酸酶(Arg-1)的活力,肝脏中丙二醛(MDA)的含量和超氧化物歧化酶(SOD)的活力,并进行肝脏病理组织学检查。结果显示葛根护肝片可降低慢性酒精性肝损伤大鼠血清中ALT、PNP、α-GST、GLDH和Arg-1的活力,可降低血清中TC、TG和LDL-C的含量,减少肝脏中的MDA含量,减少肝脏脂肪变性程度。说明葛根护肝片对慢性酒精性肝损伤模型大鼠有保护作用。
To study the protective effect of Radix Puerariae liver-protecting tablet(RPLT)on chronic alcoholic liver injury in rats.The model rats of chronic alcoholic liver injury was prepared by increasing the intragastric concentration of alcohol by stages in 4 weeks.The model rats were randomly divided into model group,positive drug group(333.3 mg·kg^(-1)),high(520.8 mg·kg^(-1)),medium(260.4 mg·kg^(-1)),a nd low(130.2 mg·kg^(-1))dose group,other normal rats were set as blank control group.All the subjects were given four weeks consecutively.The activity of ALT,AST,PNP,GLDH,α-GST,PNP,Arg-1 and content of TBIL,TC,TG,LDL-C in serum was determined after the last intragastric administration.The content of MDA and activity of SOD in liver and histopathological examination of liver was determined.The result showed that RPLT can reduce the activity of ALT,PNP,α-GST,GLDH and Arg-1 in serum of rats with chronic alcoholic liver injury,reduce the content of TC,TG and LDL-C in serum,reduce the content of MDA in liver,and reduce the degree of liver steatosis.It was concluded RPLT has protective effect on chronic alcoholic liver injury model rats.
