摘要 为了揭示核桃Alfin-like家族在核桃响应低温胁迫中的作用,本研究通过生物信息学与RNA-seq结合的方法,在核桃全基因组数据库搜索Alfin-like家族基因,对该家族进行生物信息学和转录表达分析。结果表明,核桃Alfin-like基因家族包含8个成员,分布在5条染色体上,蛋白长度范围为240(JrAL5)~254(JrAL1)个氨基酸,开放阅读框为723(JrAL5)~771 bp(JrAL3),等电点为5.04~5.71,全部为酸性不稳定蛋白;含有典型的DUF3594 Domian和PHD domian 2个高度保守的结构域,聚类分析发现,该类基因可与双子叶植物拟南芥较近,与单子叶植物水稻较远。在低温胁迫48 h后经转录表达分析,JrAL2为差异表达基因,并且在种仁和芽中表达较高,在老叶和雄花中较低,推测JrAL2可能响应低温胁迫。 In order to reveal the role of walnut Alfin-like family in walnut response to low temperature stress, this study used the method of combining bioinformatics and RNA-seq to search for Alfin-like family genes in the walnut genome database, and conducted bioinformatics and transcription expression analysis on the family. The results showed that the walnut Alfin-like gene family contains 8 members, distributed on 5 chromosomes, the protein length ranges from 240(JrAL5) to 254(JrAL1) amino acids, the open reading frame is 723(JrAL5) to 771 bp(JrAL3), and the isoelectric point is 5.04-5.71, all of which are acid-labile proteins. They contain two highly conserved domains of typical DUF3594 domian and PHD domain. Cluster analysis found that this type of gene can be compared with the dicotyledon Arabidopsis thaliana and has farther relationship with monocotyledonous rice. After 48 h of low temperature stress, the transcriptional expression analysis showed that Jr AL2 is a differentially expressed gene, and its expression is higher in seeds and buds, but lower in old leaves and male flowers. It is inferred that JrAL2 may respond to low temperature stress. The results of this study will provide important clues and basis for further research on the functions of such genes.
