摘要
口腔细胞样本和血液样本是法医DNA检验与数据库建设常见(用)的样本类型,FTA卡以其能够抑制微生物生长、抗紫外线照射的功能而常被选用于这两类样本的保存。本文报道收集上述两类样品采集卡:6种针对口腔细胞样本(含FTA指示卡)、12种用于血液样本(含FTA卡),就它们样本保存区域的属性、抑制微生物、抗紫外线照射以及PCR扩增效果等进行系统分析。结果发现只有具有化学物质涂层的采集卡才有有效抑制微生物生长、抗紫外线照射的功能;而具有涂层的采集卡则因涂层中存在的化学物质会对PCR扩增效果略有影响。基于以上分析,本文认为具有抑制微生物生长、抗紫外线照射功能的口腔细胞样本和/或血液样本采集卡更适合法医DNA检验与数据库样本的保存及STR检测。
Saliva(usually harboring buccal cells)and blood samples are among commonly-used evidential materials for DNA detection and database construction.FTA card,boasted of ability to inhibit microbial growth and resist ultraviolet radiation,is therewith frequently selected to preserve oral cells and/or blood samples.With collection of six kinds of saliva sample acquisition card(including FTA indicator card)and twelve ones for blood sample(FTA card enclosed),this paper systematically analyzed these cards about their properties of sample-storage area,microorganism inhibition,resistance against ultraviolet radiation and effect on PCR amplification.It revealed that only the chemical-processing cards were able to effectively inhibit the growth of microorganisms.Moreover,the cards owning coating layer can slightly affect PCR amplification with the presence of chemical substances in coating layer.Therefore,suggestions are kindly given that the appropriate selection should be placed on the sample acquisition cards that are functional of inhibiting the growth of microorganisms and resisting against ultraviolet radiation.
