丹酚酸B通过Nrf2通路改善CCl_(4)介导的大鼠肾损伤 被引量:1

摘要 目的:探讨丹酚酸B(SalB)通过核因子E2相关因子2(Nrf2)通路改善CCl_(4)介导的大鼠肾组织氧化应激状态。方法:采用50%CCl_(4)腹腔注射建立AKI大鼠模型,并对模型组大鼠进行SalB(80、160 mg/kg)灌胃治疗,检测血清肌酐(SCr)、尿素氮(BUN)水平,采用HE染色进行病理检测;比色法检测肾组织中丙二醛(MDA)、总超氧化物歧化酶(T-SOD)的水平;荧光定量PCR检测Nrf2、HO-1的mRNA表达水平。结果:通过HE验证成功制备AKI模型;血清学结果表明SCr、BUN在CCl_(4)模型组升高(P<0.05),在SalB治疗组下降(P<0.05);在CCl_(4)模型组,SOD含量降低,MDA含量升高(P<0.05),在SalB治疗组SOD含量升高,MDA含量下降(P<0.05);荧光定量PCR结果显示CCl_(4)模型组肾组织中Nrf2和HO-1表达降低(P<0.05);SalB能增加CCl_(4)模型组肾组织中Nrf2和HO-1的表达水平(P<0.05)。结论:SalB能通过Nrf2通路改善CCl_(4)所介导的大鼠肾组织氧化应激状态。 Objective:To investigate the effect of salvianolic acid B(SalB)on CCl_(4) mediated oxidative stress in rat kidney through Nrf2 pathway.Methods:AKI rat model was established by intraperitoneal injection of 50%CCl_(4),and then subjected to intragastric administration with SalB in dose of 80 mg/kg and 160 mg/kg,respectively.Levels of serum creatinine(SCr)and urea nitrogen(BUN)were detected,and the kidney tissues were taken and undergone pathological examination via HE staining.Colorimetry was used to measure the levels of malondialdehyde(MDA)and total superoxide dismutase(T-SOD)in renal tissues.Fluorescent quantitative PCRwas performed to detect mRNA expression level of Nrf2 and HO-1.Results:AKI model was successfully prepared by HE validation;Serological measurement showed that SCr and BUN were increased in CCl_(4) model group(P<0.05),yet decreased in SalB treatment group(P<0.05).Decreased SOD content and increased MDA level were observed in CCl_(4) model group(P<0.05).SOD content was increased,and MDA level was decreased in SalB treatment group(P<0.05).Fluorescence quantitative PCRresults showed that the expression of Nrf2 and HO-1 was decreased in CCl_(4) model group,and SalB facilitated up-regulation of Nrf2 and HO-1 expression in renal tissues from rats in CCl_(4) model group(P<0.05).Conclusion:SalB can improve CCl_(4) mediated oxidative stress in rat kidney through Nrf2 pathway.
