野木瓜渣膳食纤维改性工艺优化及功效研究

Optimization and Efficacy of Modification of Stanuntonia Chinensis Pomace Dietary Fiber

摘要 为探索野木瓜渣膳食纤维高值化改性,提高可溶性膳食纤维(SDF)和总膳食纤维(TDF)得率,通过单因素试验和响应面优化,得到野木瓜渣膳食纤维H_(2)O_(2)法改性最优工艺为:H_(2)O_(2)浓度2.15%、温度60.7 ℃、时间2.5 h、pH9.0,此条件下SDF得率为20.57%,TDF得率达到79.24%;酶解法改性最优工艺为:料液比1:21.8、纤维素酶量2.9%、温度45.5 ℃、时间3 h,此条件下SDF得率为27.41%,TDF得率为84.17%。酶解法改性的SDF和TDF得率均高于H_(2)O_(2)法改性(P<0.01),酶解法改性多酚和黄酮的保全率远高于H_(2)O_(2)法改性(P<0.01),且持水力、持油力和膨胀力等物理性质和对亚硝酸盐、胆固醇和阳离子的吸附能力均明显优于H_(2)O_(2)法改性(P<0.01),对胆酸盐吸附能力弱于H_(2)O_(2)法改性(P<0.01)。酶解法改性总体优于H_(2)O_(2)法改性。 Using the soluble dietary fiber (SDF) yield and total dietary fiber (TDF) yield as index,the optimum technology for dietary fiber from the Stanuntonia chinensis pomace (SCP) was obtained by H_(2)O_(2) method and enzymatic method.Both methods were optimized by single factor tests and Box-Behnken Design response surface optimization.The results showed that the optimal conditions of H_(2)O_(2) method was as follows:H_(2)O_(2) concentration 2.15%,temperature 60.7 ℃,reaction time 2.5 h and pH 9,under this condition,the SDF and TDF yield was 20.57% and 79.24%.The optimal conditions of enzymatic method was:solid-liquid ratio 1:21.8 g/mL,cellulase concentration 2.9%,temperature 45.5 ℃ and enzymatic time 3 h,under this condition,the SDF and TDF yield was 27.41% and 84.17% respectively.The polyphenol and flavone preservation rate of enzymatic modification was 82.72% and 53.37% respectively,far higher than that of H_(2)O_(2) modification (P<0.01).The water-holding capacity,oil-holding capacity,expansion force and the nitrite,cholesterol,cation adsorption ability of the TDF by enzymatic method were significantly better than those of H_(2)O_(2) method (P<0.01).The enzymatic method was better than the H_(2)O_(2) method not only in increasing the yield of the SDF and TDF,but also in preservation rate of the SCP characteristic ingredients.
