三七花活性成分提取工艺优化及其组成成分分析 被引量:4

Extraction Process Optimization and Analysis of Active Components from Panax Notoginseng Flowers

摘要 为促进三七花的深度开发利用。以单因素-响应面法优化三七花活性成分的浸提条件,利用紫外分光光度法、液相色谱质谱联用法(LC-MS)、高效液相色谱法(HPLC)测定三七花及其提取物中皂苷、多酚及氨基酸的组成及含量。以21.8倍52.9%体积分数乙醇常温浸提50.8 h,浸提2次,120 ℃喷雾干燥得到水溶性好、汤色黄绿、明亮、花香浓郁、微苦回甘的三七花提取物,得粉率(46.63±0.81)%,总皂苷含量(419.39±0.03)mg/g;LC-MS检测到皂苷等39种活性成分,HPLC测定了14种皂苷、10种多酚及17种氨基酸的含量,以人参皂苷F1((107.67±1.04)mg/g)、Rb2((55.57±0.25)mg/g)、Rb3((59.09±2.73)mg/g)和儿茶素((50.25±0.68)mg/g)、没食子酸((6.21±0.17)mg/g)、没食子酸儿茶素((4.20±0.1)mg/g)等活性成分含量较高;富含GABA((19.85±1.54)mg/g)、甘氨酸((9.56±0.51)mg/g)、丝氨酸((5.23±0.36)mg/g)等营养成分,农残重金属达标。以三七花为原料开发了富含活性物质与营养成分、滋味较好的提取物,为三七花的深度开发提供了试验依据。 In order to promote the deep development and utilization of Panax notoginseng flowers.The extraction conditions of active components from Panax notoginseng flowers were optimized by single factor-response surface methodology.The components and contents of saponins,polyphenols and amino acids in Panax notoginseng flowers and their extracts were determined by ultraviolet spectrophotometry,liquid chromatography-mass spectrometry (LC-MS) and high performance liquid chromatography (HPLC).21.8 times 52.9% volume fraction of ethanol was used to extract at normal atmospheric temperature for 50.8 h,extracted for 2 times,and spray dried at 120 ℃ to obtain their extracts with a rate of (46.63±0.81)%.The extracts have good water-soluble,the color was yellow-green,bright,flowery and slightly bitter with post sweet.The total saponins were (419.39±0.03)mg/g.There were 14 saponins,10 polyphenols and 17 amino acids were determined by HPLC.Such as ginsenoside F1 ((107.67±1.04)mg/g),Rb2 ((55.57±0.25)mg/g),Rb3 ((59.09±2.73)mg/g),catechin ((50.25±0.68) mg/g),gallic acid ((6.21±0.17)mg/g),catechin gallate ((4.20±0.1)mg/g) were full in Panax notoginseng flowers extracts,and been rich with GABA ((19.85±1.54)mg/g),glycine ((9.56±0.51)mg/g),serine ((5.23±0.36)mg/g) and other nutrients.The extracts with Panax notoginseng flowers as raw materials which were rich in active substances and nutrients with good tastes were developed.It was provided an experimental basis for the further development of Panax notoginseng flowers.
