胃癌组织CDK2、GDF-11的表达及其与幽门螺旋杆菌感染的关系 被引量:3

The expression of CDK2 and GDF-11 in gastric cancer tissues and their relationship with Helicobacter pylori infection

摘要 目的检测胃癌组织中细胞周期蛋白依赖性激酶2(CDK2)、生长分化因子11(GDF11)表达情况,探讨二者与胃癌患者幽门螺旋杆菌(HP)感染的关系。方法 2018年1月-2020年5月在本院就诊并接受手术治疗的胃癌患者91例,手术过程中取胃癌组织与正常胃黏膜组织,采用免疫组织化学染色检测CDK2、GDF11蛋白水平,分析二者与胃癌临床病理特征的关系及与HP感染的相关性。结果 91例胃癌患者中,快速尿素酶试验检出HP阳性66例,Giemsa染色检查HP阳性58例,合计检出HP感染75例。免疫组织化学染色检查CDK2主要定位于胃癌组织中的细胞核与细胞质,GDF11主要定位于细胞质与细胞膜,胃癌组织CDK2(63.74%)、GDF11(72.53%)蛋白阳性率显著高于正常胃黏膜组织(27.47%、24.18%)(均P<0.01)。胃癌组织CDK2、GDF11的表达与肿瘤分化程度、淋巴结转移、临床分期及浸润深度有关(P<0.05或P<0.01),与性别、年龄、肿瘤直径和病理分型无显著相关性(均P>0.05)。HP感染胃癌组织CDK2、GDF11阳性率分别为73.33%(55/75)和80.00%(60/75),与未感染HP胃癌组织CDK2、GDF11阳性率18.75%(3/16)和37.5%(6/16)比较差异均有统计学意义(均P<0.01)。结论胃癌组织CDK2、GDF-11高表达,且二者与肿瘤的分化程度、淋巴结转移、临床分期及HP感染有关。 Objective To detect the expression of cyclin-dependent kinase 2(CDK2) and growth differentiation factor 11(GDF11) in gastric cancer tissues and to explore the relationship between the two and Helicobacter pylori infection in patients with gastric cancer. Methods Subjects were 91 patients with gastric cancer who underwent surgery at this Hospital from January 2018 to May 2020. Samples of gastric cancer tissue and normal gastric mucosal tissue were collected during surgery, and the level of expression of the proteins CDK2 and GDF11 was determined using immunohistochemical staining. The relationship between the two and the clinicopathological characteristics of gastric cancer and their correlation with H. pylori infection were analyzed. Results Of 91 patients with gastric cancer, 66 were positive for H. pylori according to a rapid urease test, 58 were positive for H. pylori according to Giemsa staining, and a total of 75 were infected with H. pylori. Immunohistochemical staining indicated that CDK2 was mainly located in the nucleus and cytoplasm of gastric cancer tissues, and GDF11 was mainly located in the cytoplasm and cell membranes. Positivity for the CDK2 protein(63.74%) and the GDF11 protein(72.53%) in gastric cancer tissue was markedly higher than that in normal gastric mucosa tissue(27.47%, 24.18%, P<0.01 for both). The expression of CDK2 and GDF11 in gastric cancer tissue is related to tumor differentiation, lymph node metastasis, clinical stage, and depth of invasion(P<0.05 or P<0.01 for all) and is not significantly correlated with sex, age, tumor size, or pathological type(P>0.05 for all). In gastric cancer tissue infected with H. pylori, positivity for CDK2 was 73.33%(55/75) and positivity for GDF11 was 80.00%(60/75) compared to positivity for CDK2 of 18.75%(3/16) and positivity for GDF11 of 37.5%(6/16) in gastric cancer tissue not infected with H. pylori. The positivity of those proteins differed significantly(P<0.01 for both). Conclusion CDK2 and GDF-11 are highly expressed in gastric cancer tissues, and both are related to clinicopathological characteristics such as tumor differentiation, lymph node metastasis, clinical stage, and H. pylori infection in gastric cancer tissues.
