摘要
为了研究幼年与成年湖羊的超数排卵及卵巢基因表达差异,试验以4周龄幼年母羊(Lamb组)和2岁龄成年母羊(Adult组)湖羊为研究对象,对其进行超数排卵处理,观测试验羊卵巢的表型差异;采用放射免疫法测定试验羊血清繁殖激素水平;以实时荧光定量PCR法测定试验羊卵巢组织CYP17A1、INHA和FSHR基因相对表达量。结果表明:超数排卵处理后,Lamb组母羊卵巢形成的卵泡数量为(70.40±6.11)个,显著高于Adult组的(21.40±2.87)个(P<0.05);Adult组母羊卵细胞直径和发情率显著高于Lamb组(P<0.05)。Adult组母羊血清繁殖激素水平与Lamb组比较差异不显著(P>0.05)。Lamb组母羊卵巢组织CYP17A1、INHA和FSHR基因的相对表达量均极显著高于Adult组(P<0.01)。说明幼年母羊通过提高卵巢组织内与雌激素合成相关的基因表达水平,而使其对外源超数排卵激素较敏感。
In order to study the superovulation and the difference of ovarian gene expression between juvenile and adult Hu sheep, 4-week-old young ewes(Lamb group) and 2-year-old adult ewes(Adult group) Hu sheep were selected as the research subjects, which were treated with superovulation, and the phenotypic difference of ovary was observed by surgical operation. The levels of reproductive hormone in serum of experimental sheep were measured by radioimmunoassay. The relative expression levels of CYP17 A1, INHA and FSHR genes were determined by real-time fluorescent quantitative PCR. The results showed that the number of ovarian follicles in Lamb group after superovulation was(70.40±6.11), which was significantly higher than that in Adult group(21.40±2.87, P<0.05). The follicle diameter and estrus rate in Adult group were significantly higher than those in Lamb group(P<0.05). There was no significant difference in serum reproductive hormone level between the Adult group and the Lamb group(P>0.05). The expression levels of CYP17 A1, INHA and FSHR in Lamb group were extremely significantly higher than those in Adult group(P<0.01). It shows that young ewes are more sensitive to exogenous hormones by increasing the expression level of estrogen related genes in ovarian tissue.
