摘要
目的选择合适的消毒剂在法医DNA实验室进行DNA污染清除和DNA无损提取两个方向进行应用。方法使用4种消毒剂作用于9947、口腔卡、不同浓度血液,然后进行相应的DNA模板制备和检测。结果使用复合季铵盐类消毒剂作用20 min,对9947、口腔卡以及20倍稀释血液中的DNA呈现出明显的清除作用,对5倍稀释血液中DNA的清除作用减弱。使用PCMX消毒剂作用20 min,对9947、口腔卡以及不同浓度血液中的DNA均未呈现出明显的清除作用。结论复合季铵盐类消毒剂可以进行DNA污染清除,PCMX消毒剂不会影响DNA的检测和提取。
Objective To select a suitable disinfectant for application in the forensic DNA laboratory to remove DNA contamination and extract intact DNA.Methods Four kinds of disinfectants were used to treat 9947,buccal card and blood with different concentrations,and then the corresponding DNA templates were prepared and detected.Results When compound quaternary ammonium salt disinfectant was used for 20 min,it showed obvious removal effect on the DNA in 9947,buccal card and 20-fold diluted blood,and had a weakened removal effect on the DNA in 5-fold diluted blood.PCMX disinfectant was use for 20 min,but it showed no obvious removal effect on the DNA in 9947,buccal card and blood with different concentrations.Conclusion The compound quaternary ammonium salt disinfectant has the ability to remove DNA contamination,while the PCMX disinfectant will not affect the detection and extraction of DNA.
