线栓法制备大脑中动脉闭塞大鼠模型问题探析 被引量:2

摘要 目的:分析影响线栓法制备大脑中动脉闭塞(middle cerebral artery occlusion, MCAO)大鼠模型成功率的因素。方法:通过实践探究线栓法制备MCAO模型过程中遇到的问题,包括大鼠的品系、体质量、性别;造模操作的术前准备:麻醉剂、手术切口、线栓的选择,线栓入颅的部位及深度、术中和术后应该注意的事项及处理方式等;采用文献检索及自身认识对问题进行分析。结果:制备模型过程中,其存活率因不同操作过程而偏差极大,大鼠品种及体质量的选择、麻醉剂选用、线栓处理及入颅深度、术后护理等因素均能对模型成功率产生影响。结论:提高造模者的手术熟练程度并对大鼠进行术后护理,可提高模型制备的成功率。 Objective:To analyze the factors influencing the success rate of middle cerebral artery occlusion(MCAO)model in rats.Methods:The problems encountered in the process of making MCAO model by thread blot method,including rat strain,gender,preoperative preparation of modeling operation,selection of anesthetics,surgical incision and thread blot,location of thread blot into skull,matters needing attention during and after operation,and treatment methods,etc.were explored,then the relevant problems by literature search and self-knowledge were analyzed.Results:In the process of preparing the model,the survival rate had great deviation due to the influence of different operation processes.The selection of rat species and body weight,the selection of anesthetics,the treatment of thread bolt and the depth of craniotomy,postoperative nursing and other factors can affect the success rate of the model.Conclusion:The success rate of model preparation can be improved by improving the operation proficiency of the model maker and postoperative nursing of rats.
