摘要
目的:分析柴胡皂苷d抗肝纤维化模型中的大鼠脂质过氧化作用。方法:将24例SPF级雄性SD大鼠随机分为三组,即预防组、模型组、正常组,每组8例。统计分析三组大鼠的血清酶活性、血清透明质酸(HA)、层粘连蛋白(LN)、Ⅳ型胶原(IV-C)水平及血清、肝组织超氧化物歧化酶(SOD)、丙二醛(MDA)水平、肝纤维化程度、胶原总面积、面积比。结果:模型组、正常组、预防组大鼠的血清谷草转氨酶(AST)、佛甲草提取液(SLT)水平均逐渐降低(P<0.05),模型组、预防组、正常组大鼠的血清HA、LN、IV-C水平均逐渐降低(P<0.05)。模型组、预防组大鼠的血清SOD水平均低于正常组(P<0.05);模型组、正常组、预防组大鼠的血清、肝组织MDA水平逐渐降低(P<0.05),肝组织SOD水平逐渐升高(P<0.05)。模型组、预防组大鼠的肝纤维化0级比例均低于正常组(P<0.05);预防组大鼠的肝纤维化Ⅱ级比例高于模型组、正常组(P<0.05);模型组大鼠的肝纤维化Ⅲ级比例高于正常组、预防组(P<0.05)。模型组、预防组、正常组大鼠的胶原总面积、面积比均逐渐降低(P<0.05)。结论:柴胡皂苷d抗肝纤维化机制可能和抗脂质过氧化作用相关。
Objective:To analyze the effect of saikosaponin d on lipid peroxidation in rats with hepatic fibrosis.Methods:24 SPF male SD rats were randomly divided into three groups:prevention group,model group and normal group,with 8 cases in each group.The levels of serum enzyme activity,serum hyaluronic acid(HA),laminin(LN),type Ⅳ collagen(Ⅳ-C),serum and liver superoxide dismutase(SOD),malondialdehyde(MDA),degree of liver fibrosis,total area and area ratio of collagen were statistically analyzed.Results:The levels of serum aspartate aminotransferase(AST)and SLT in model group,normal group and prevention group decreased gradually(P<0.05),and the levels of serum HA,LN and Ⅳ-C in model group,prevention group and normal group decreased gradually(P<0.05).The levels of serum SOD in model group and prevention group were lower than those in normal group(P<0.05);The levels of MDA in serum and liver tissue of model group,normal group and prevention group decreased gradually(P<0.05),and the level of SOD in liver tissue increased gradually(P<0.05).The proportion of grade 0 liver fibrosis in model group and prevention group was lower than that in normal group(P<0.05);The proportion of grade Ⅱ liver fibrosis in the prevention group was higher than that in the model group and normal group(P<0.05);The proportion of grade Ⅲ liver fibrosis in model group was higher than that in normal group and prevention group(P<0.05).The total area and area ratio of collagen in model group,prevention group and normal group decreased gradually(P<0.05).Conclusion:The anti fibrosis mechanism of saikosaponin d may be related to its anti lipid peroxidation effect.
