链脲佐菌素联合高脂饮食制备糖尿病小鼠模型研究 被引量:3

A study on the preparation of diabetic mouse model with streptozotocin and high fat diet

摘要 采用高脂饲料、腹腔注射链脲佐菌素(STZ)、高脂饲料+腹腔注射STZ等3种方法分别制备2型糖尿病小鼠模型,通过检测小鼠血糖水平和糖化血红蛋白(HbA1c)水平,评估所建模型是否成功。高脂饲料组小鼠第7周时血糖水平显著升高,而HbA1c水平与正常对照组无差异;腹腔注射STZ组小鼠血糖水平仅一过性升高后即恢复正常,HbA1c水平与正常对照组无差异;高脂饲料+腹腔注射STZ组小鼠第2周起血糖水平明显高于正常对照组,第3周起HbA1c水平明显高于正常对照组,随后逐渐升高,并维持高水平。采用高脂饲料+腹腔注射STZ法构建2型糖尿病小鼠模型具有省时、简便、经济等优点。 Type II diabetic mouse models were prepared by 3 different methods including high-fat diet,intraperitoneal injection of streptozotocin(STZ),high-fat diet and intraperitoneal injection of STZ.The blood glucose level and glycosylated hemoglobin(HbA1c)level of mice were detected to evaluate whether the model was successful.The blood glucose level of mice in the high-fat diet group increased significantly at the 7th week,but there was no difference in HbA1c level between the high-fat diet group and the normal control group.The blood glucose level of mice in the intraperitoneal injection STZ group returned to normal only after a transient increase,and there was no difference in HbA1c level between the STZ group and the normal control group.The blood glucose level of mice in the high-fat diet+intraperitoneal injection STZ group was significantly higher than that of the normal control group from the second week,while the HbA1c level was significantly higher than that of the normal control group from the third week,and then gradually increased and maintained a high level.The use of high-fat diet plus intraperitoneal injection of STZ to build a diabetic mouse model is time-saving,simple and economical.
