葛根提取物对CCl_(4)致小鼠急性肝损伤的保护作用

Protective Effect of Pueraria Lobata Extract on CCl_(4)-induced Liver Eamage in Mice

目的:探讨葛根提取物对四氯化碳(CCl_(4))致小鼠急性肝损伤的保护作用。方法:将60只昆明小鼠随机分为正常对照组、模型组,葛根提取物高、中、低剂量组(400、200、100 mg/kg),联苯双酯组(100 mg/kg)。连续灌胃给药14 d后腹腔注射0.2%CCl_(4)溶液诱发急性肝损伤,24 h后处死小鼠,检测血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆汁酸(TBA)、总胆红素(TBIL)、碱性磷酸酶(ALP)、γ-谷氨酰转移酶(γ-GT)含量和肝组织中总超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px),总抗氧化能力(T-AOC)、丙二醛(MDA)、白介素-1β(IL-1β)、白介素-6(IL-6)、转化生长因子-β(TGF-β)、肿瘤坏死因子-α(TNF-α)含量。结果:与模型组比较,各给药组小鼠ALT、AST、TBA、TBIL、ALP、γ-GT、MDA、IL-1β、IL-6、TGF-β、TNF-α的含量降低(P<0.05或0.01),SOD、T-AOC、GSH-Px含量升高(P<0.05或0.01)。结论:葛根提取物对CCl_(4)致小鼠急性肝损伤具有较好的保护作用,其机制可能与增强抗氧化能力及抗炎作用相关。

Objective To explore the effect of Pueraria lobata extractt on CCl_(4)-induced liver damage in mice.Methods 60 mice were randomly divided into normal control group,model group,Pueraria lobata extract high,medium and low dose groups(400,200,100 mg/kg),and bifendate group(100 mg/kg).After continuous intragastric administration for 14 days.Acute liver injury model was established by intraperitoneal injection of 0.2%CCl_(4) olive oil solution.After 24 hours of modeling,the contents of alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bile acid(TBA),total bilirubin(TBIL),alkaline phosphatase(ALP),gamma glutamyltransferase(γ-GT)in serum and total superoxide dismutase(SOD),malondialdehyde(MDA),total antioxidant capacity(T-AOC)glutathione peroxidase(GSH-Px),interleukin-1β(IL-1β),interleukin-6(IL-6),transforming growth factor-β(TGF-β)and tumor necrosis factor-α(TNF-α)in liver tissue were detected.Results Compared with the group,pueraria extract significantly reduced ALT、AST、TBA、TBIL、ALP、γ-GT、MDA、IL-1β、IL-6、TGF-βand TNF-αlevels in mice with acute liver injury(P<0.05 or 0.01),significantly increased SOD,T-AOC,GSH-Px levels(P<0.05 or 0.01).Conclusion Pueraria lobata extract has a significant protective effect against acute liver injury induced by CCl_(4) in mice,the mechanism may be related to the enhancement of antioxidant capacity and anti-inflammatory effect.
