摘要
目的采用离子迁移谱(ion mobility spectrometry,IMS)技术,建立中成药和保健食品中快速筛查二甲双胍、苯乙双胍、罗格列酮和吡格列酮的方法。方法设定条件为正离子源检测,离子源电压1800 V、漂移管电压7500 V、进气温度180℃、漂移管温度180℃、栅电压45 V、栅脉冲宽度120μs、漂移管流速1.2 L·min^(-1)、排气泵0.8 L·min^(-1)、运行时间30 s、光谱范围25 ms,样品用甲醇直接提取,稀释后直接进样,与对照品的迁移时间进行比对分析,判断是否有降糖类化合物的添加。结果可以实现4种化合物的单一成分或多成分同时存在非法添加的检测,根据信噪比能确定4种非法添加物质的定量限和检出限。结论建立方法简便、快速、重现性好,可以用于中成药和保健食品中非法添加二甲双胍、苯乙双胍、罗格列酮和吡格列酮的初步筛查。
Objective To establish a rapid screening method for metformin,phenformin,rosiglitazone and pioglitazone in traditional Chinese patent medicines and health care products by ion mobility spectroscopy(IMS).Methods The conditions were set as positive ion source detection,ion source voltage 1800 V,drift tube voltage 7500 V,intake temperature 180℃,drift tube temperature 180℃,gate voltage 45 V,gate pulse width 120μs,drift tube flow rate 1.2 L·min^(-1),exhaust pump 0.8 L·min^(-1),operation time 30 s,spectral range 25 ms.The sample is extracted directly with methanol,then injected directly after dilution.The migration time of the sample was compared with that of the control sample to determine whether there was the addition of hypoglycemic compounds.Results The detection of illegal addition of single or multi-component of four compounds can be realized.The quantitative limit and detection limit of four illegal addition substances can be determined according to the signal-to-noise ratio.Conclusion The method was simple,rapid and reproducible.It can be used for the preliminary screening of metformin,phenformin,rosiglitazone and pioglitazone illegally added in Chinese patent medicines and health care products.
