摘要
目的:探讨六月青醇提物凝胶剂对大鼠慢性湿疹的治疗效果。方法:建立大鼠湿疹模型,随机分为模型组、阳性组及六月青醇提物凝胶剂高、中、低剂量组,每组6只,另设空白组6只。给药组给予相应药物治疗,模型组给予相应空白凝胶,每天2次,连续给药10 d,空白组不做处理。治疗过程中对大鼠背部造模区域皮损程度进行评分,实验结束后,苏木精—伊红(HE)染色观察皮肤组织病理变化,酶联免疫吸附试验(ELISA)法检测大鼠血清中白细胞介素-4(IL-4)和干扰素-γ(IFN-γ)的含量。结果:与模型组比较,六月青醇提物凝胶剂可不同程度改善大鼠湿疹模型表皮角化,减轻炎性细胞浸润及细胞间水肿,各给药组IFN-γ和IFN-γ/IL-4显著升高(P<0.05),但IL-4的含量无明显差异(P>0.05)。结论:六月青醇提物凝胶剂可明显提高湿疹大鼠血清中IFN-γ含量水平,矫正Th1/Th2失衡,改善大鼠慢性湿疹的病理变化,对湿疹具有一定的治疗作用。
Objective: To investigate the therapeutic effect of Liuyueqing alcohol extract gel on chronic eczema in rats. Methods: A rat eczema model was established and randomly divided into a model group, a positive group and Liuyueqing alcohol extract gel high-, medium-and low-dose groups, with 6 rats in each group. Six healthy rats were defined as a blank group. The drug group rats were given the corresponding drug treatment, and ratsin the model group were given the blank gel twice a day for 10 consecutive days. Rats in blank group did not receive any treatment. During the treatment, the degree of skin lesions on the back modeling area of rat was scored.After experiment, HE staining was used to observe the pathological changes of the skin tissue. The contents of interleukin-4(IL-4) and interferon-γ(IFN-γ) in the serum were detected by enzyme-linked immunosorbent assay(ELISA). Results: Liuyueqing alcohol extract gel could improve the epidermal keratinization, reduce inflammatory cell infiltration and attenuate intercellular edema ineczema ratsin different degrees. The IFN-γ level and IFN-γ/IL-4 ratio were increased significantly in all Liuyueqing administration groups compared with the model group(P<0.05), but there was no significant difference in IL-4 content(P>0.05). Conclusion: Liuyueqing alcohol extract gel can significantly increase the serum IFN-γ level, correct the imbalance of Th1/Th2, and improve the pathological changes of chronic eczema in rats. It has a certain therapeutic effect on eczema.
