摘要
目的利用生物信息学,研究CXCL13在非小细胞肺癌(NSCLC)中的作用。方法利用Gepia数据库分析CXCL13在NSCLC患者中mRNA表达。利用Proteinatlas数据库分析CXCL13在NSCLC患者肺中蛋白表达水平并对肺腺癌(LUAD)与肺鳞癌(LUSC)中检测进行比较。通过网页工具TIMER研究CXCL13在NSCLC患者免疫浸润中的作用。CancerSEA分析CXCL13在不同GEO数据集中表达。利用Ualcan数据库分析CXCL13在NSCLC中启动子甲基化情况。结果Gepia结果显示CXCL13在NSCLC中mRNA的表达明显高于正常组。Proteinatlas结果显示NSCLC癌组织中CXCL13蛋白表达高于正常肺组织。TIMER分析结果显示CXCL13的表达在NSCLC患者中肿瘤纯度呈负相关,与NSCLC患者的B细胞、T细胞CD8^(+)细胞、Tregs细胞浸润呈正相关(P<0.05),但相关程度比较低(Rho<0.7)。经多因素Cox回归分析,CXCL13高表达合并高水平T细胞CD8^(+)细胞浸润组LUSC患者预后好于CXCL13高表达合并低水平T细胞CD8^(+)细胞浸润组(P<0.05),CXCL13高表达合并低水平Tregs细胞浸润组LUSC患者预后好于CXCL13高表达合并高水平Tregs细胞浸润组(P<0.05)。CancerSEA分析显示CXCL13表达在NSCLC与转移和侵袭呈正相关,与DNA修复呈负相关。Ualcan分析显示CXCL13在LUAD患者中启动子甲基化呈高甲基化状态,在LUSC患者中呈低甲基化状态。结论CXCL13参与NSCLC免疫细胞浸润和肿瘤细胞功能,CXCL13也许可以作为NSCLC免疫治疗的靶点之一。
Objective To investigate the role of CXCL13 in non-small cell lung cancer(NSCLC)using bioinformatics.Methods The mRNA expression of CXCL13 in NSCLC patients was analyzed by Gepia data base.The protein expression level of CXCL13 in the lungs of NSCLC patients was analyzed by Proteinatlas data base.The effect of CXCL13 on immune infiltration in NSCLC patients was investigated by using web tool TIMER.The expression of CXCL13 in different GEO databases was analyzed by CancerSEA.The promoter methylation of CXCL13 in NSCLC was analyzed by Ualcan data base,comparison was evaluated in lung adenocarcinoma(LUAD)and lung squamous carcinoma(LUSC).Results Gepia results showed that the mRNA expression of CXCL13 in NSCLC was significantly higher than that in the normal group.Proteinatlas results showed that CXCL13 protein expression in NSCLC cancer tissues was higher than that in normal lung tissues.TIMER analysis showed that CXCL13 expression was negatively correlated with tumor purity in NSCLC patients,and positively correlated with invasion of B cells,T cell CD8^(+)cells and Tregs cells(P<0.05),but the degree of correlation was low(Rho<0.7).Multivariate Cox regression analysis showed that the prognosis of LUSC patients in CXCL13 high expression group with high level T cell CD8^(+)invasion group was better than that in CXCL13 high expression group with low level T cell CD8^(+)invasion group(P<0.05).The prognosis of LUSC patients in CXCL13 high expression group with low Tregs cell invasion was better than that in CXCL13 high expression group with high Tregs cell invasion group(P<0.05).CancerSEA analysis showed that CXCL13 expression in NSCLC was positively correlated with metastasis and invasion,and negatively correlated with DNA repair.Ualcan analysis showed that CXCL13 promoter methylation was hypermethylated in patients with LUAD,and hypomethylated in patients with LUSC.Conclusions CXCL13 is involved in NSCLC immune cell invasion and tumor cell function,and CXCL13 may be one of the targets of NSCLC immunotherapy.
