星形细胞瘤生存预后相关miRNA⁃mRNA调控关系对筛选 被引量:1

Screening of miRNA⁃mRNA regulatory pairs related to survival and prognosis of astrocytoma

摘要 利用GEO数据库中的芯片数据,筛选与星形细胞瘤生存预后相关的miRNA⁃mRNA调控关系对,为后续研究提供理论支持。下载芯片数据利用R语言进行差异表达分析,得到星形细胞瘤较正常组织表达显著改变的miRNA与mRNA;通过miRNA靶基因预测,将靶基因与差异表达mRNA取交集,明确mRNA与miRNA之间的关系;利用GEPIA2.0工具在TCGA数据库中筛选有生存价值的mRNA并验证表达情况,利用OncoLnc工具对相应miRNA进行生存分析。筛选到差异表达的miR⁃NA 90个(表达上调22个,下调68个);差异表达的mRNA 644个(表达下调476个,上调168个);根据miRNA靶基因预测结果,整理出miRNA⁃mRNA关系对30个,对其中的mRNA与miRNA进行生存分析,共得到7个miRNA⁃mRNA关系对与LGG患者生存预后明显相关,未筛选到与GBM患者生存预后有关的调控关系对。本研究筛选到的7个miRNA⁃mRNA调控关系对与LGG患者生存预后显著相关,可为相关研究与治疗提供靶点和参考方向。 Microarray data in the GEO database were used to screen miRNA⁃mRNA regulatory pairs related to the survival and prognosis of astrocytoma,so as to provide theoretical support for subsequent studies.The data were analyzed using R language for differential expression,and the miRNAs and mRNAs with significantly changed expression in astrocytoma compared with normal tissues were obtained.By predicting miRNA target genes,the intersection of target genes and differentially expressed mRNAs was taken to clarify the relationship between mRNA and miRNA.GEPIA2.0 tool was used to screen the mRNAs with survival value in the TCGA database and verify the expressions.OncoLnc tool was used to analyze the survival of the corresponding miRNAs.Ninety miRNAs with differential expressions were screened(22 up⁃regulated and 68 down⁃regulated).There were 644 differentially expressed mRNAs(476 down⁃regulated mRNAS and 168 up⁃regulated mRNAS).According to miRNA target gene prediction results,30 miRNA⁃mRNA regulatory pairs were sorted out,among which mRNA and miRNA were analyzed for survival.A total of seven miRNA⁃mRNA regulatory pairs were found to be significantly related to the survival prognosis of LGG patients,and no regulatory pairs related to the survival prognosis of GBM patients were screened.The seven miRNA⁃mRNA regulatory paris selected in this study were significantly correlated with the survival and prognosis of LGG patients,which can provide targets and reference directions for related research and treatment.
