瑞马唑仑诱导对脓毒症小鼠免疫和肾上腺皮质功能的影响 被引量:7

摘要 目的探讨不同麻醉药物诱导对脓毒症小鼠免疫和肾上腺皮质功能的影响。方法将BALB/C小鼠分成4组:空白对照组(N组)、脓毒症组(S组)、瑞马唑仑组(R组)及依托咪酯组(E组)。于注射后1 h和8 h各取8只小鼠摘除眼球取血,检测血浆中自然杀伤(NK)细胞、CD3^(+)、CD4^(+)、CD8^(+)的百分比,CD4^(+)/CD8^(+)及白细胞介素(IL)-6、IL-10、促肾上腺皮质激素(ACTH)和皮质酮(Cor)的含量。结果1 h时,CD3^(+)、CD4^(+)和CD8^(+)3项指标中,N组最高,S组最低,R组高于E组;CD4^(+)/CD8^(+)和NK细胞2项指标中,N组最高,S组最低,但R组与E组差别无统计学意义(P>0.05);4组间血清IL-6浓度均有差别,N组最低,S组最高,R组低于E组;4组间血清IL-10浓度均有差别,N组最高,S组最低,R组高于E组。8 h时,4组间CD4^(+)指标均有差别,N组最高,S组最低,R组低于E组;CD3^(+)、CD8^(+)和NK细胞3项指标中,N组最高,S组最低,但R组与E组的差别无统计学意义(P>0.05);N组CD4^(+)/CD8^(+)最高,S组、R组与E组的差别无统计学意义(P>0.05);4组间血清IL-6浓度有差别,N组最低,S组最高,R组与E组差别无统计学意义(P>0.05);4组间血清IL-10浓度有差别,N组最高,S组最低,R组与E组差别无统计学意义(P>0.05)。1 h和8 h时,N组ACTH和Cor含量最低,S组和R组均高于E组,而S组和R组间差别无统计学意义(P>0.05)。结论瑞马唑仑和依托咪酯单次诱导注射对脓毒症小鼠的免疫均具有保护作用,但依托咪酯抑制脓毒症小鼠的肾上腺皮质功能,而瑞马唑仑对肾上腺皮质功能无显著影响。 Objective To explore the effects of different anesthetics on the immunity and adrenal cortex function of septic mices.Methods BALB/C mices were divided into 4 groups:blank control group(group N),sepsis group(group S),remazolam group(group R)and etomidate group(group E).Eight mices in each group were taken 1h and 8h after injection to remove their eyeballs.And blood was taken to test the plasma levels of NK cells,CD3^(+),CD4^(+),CD8^(+),CD4^(+)/CD8^(+),IL-6,IL-10,ACTH and Cor.Results At 1 h,the group N was the highest,the group S was the lowest,and the group R was higher than the group E in the three indicators of CD3^(+),CD4^(+)and CD8^(+);In the two indicators of CD4^(+)/CD8^(+)and NK cells,the group N was the highest,the group S was the lowest,but there was no significant difference between the group R and the group E;IL-6 was different in the 4 groups,the group N was the lowest,the group S was the highest,the group R was lower than the group E;IL-10 was different in the 4 groups,the group N was the highest,the group S was the lowest,the group R was higher than the group E.At 8 h,the CD4^(+)was different in the 4 groups,the group N was the highest,the group S was the lowest,and the group R was lower than the group E;Among the three indexes of CD3^(+),CD8^(+)and NK cells,the group N was the highest and the group S was the lowest,but there was no significant difference between the group R and the group E;In the CD4^(+)/CD8^(+),the group N was the highest,but there was no significant difference among the group S,the group R and the group E;In IL-6,the group N was the lowest,the group S was the highest,and there were no significant difference between the group R and the group E;In IL-10,the group N was the highest,the group S was the lowest,and there were no significant difference between the group R and the group E.At 1 h and 8 h,the content of ACTH and Cor were the lowest in the group N,the group S and the group R were higher than the group E,but there were no significant difference in content of ACTH and Cor between the group S and the group R.Conclusions A single induction injection of remazolam and etomidate had a protective effect on the immunity of septic mice.However,etomidate could inhibit the adrenal cortex function of septic mice,while remazolam had no significant effect on the adrenal cortex function.
