基于生物信息学分析KNG1在胃癌中的表达变化及临床意义

摘要 目的基于生物信息学,探讨KNG1在胃癌中的表达变化及临床意义。方法利用TCGA数据库分析KNG1在胃癌中的表达及生存差异。采用Cox回归分析影响胃癌患者预后的独立危险因素,进一步纳入多因素Cox回归分析结果,构建胃癌预后列线图。运用miRWalk、TargetScanv7.2及TCGA数据库,预测调控KNG1异常表达的miRNA。通过GSEA富集分析KNG1可能参与的生物学功能。结果KNG1在胃癌组织中表达升高(P<0.01),且高表达的胃癌患者具有更差的预后(P<0.05)。多因素Cox回归分析显示,KNG1 mRNA表达、患病年龄、是否放疗及TNM分期均是影响胃癌预后的独立危险因素(P均<0.05)。纳入上述独立变量绘制的预后列线图对胃癌患者的生存时间具有较好的预测。miR-143-3p在胃癌组织中表达下调(P<0.01),与胃癌中KNG1的表达呈负相关(P<0.01)。GSEA富集分析结果显示,KNG1在胃癌中参与了mRNA分解过程中的调控、组蛋白甲基化及小RNA的产生与RNA沉默等生物学功能。结论KNG1在胃癌中表达上调,且高表达与胃癌患者不良预后相关,其可成为胃癌有前景的诊断指标、治疗靶点及预后标志物。
