癫痫患者脑脊液和血清P53 mRNA、微小RNA-34a表达情况及其与认知功能的相关性 被引量:2

Expression of P53 mRNA and microRNA-34a in cerebrospinal fluid and serum and its correlation with cognitive function in patients with epilepsy

摘要 目的探讨癫痫患者脑脊液和血清中P53 mRNA、微小RNA(miRNA)-34a的表达水平及其与认知功能的相关性。方法选取79例癫痫患者作为癫痫组,73例有头痛症状但非癫痫患者作为对照组。检测两组脑脊液和血清中P53 mRNA、miRNA-34a的表达水平并评估患者的认知功能,分析癫痫患者脑脊液和血清P53 mRNA与miRNA-34a表达水平的相关性,以及脑脊液和血清P53 mRNA、miRNA-34a相对表达水平与认知功能的相关性。结果癫痫组脑脊液P53 mRNA、miRNA-34a相对表达水平、血清P53 mRNA相对表达水平均高于对照组,血清miRNA-34a相对表达水平低于对照组(均P<0.05)。癫痫组连线测验A(TMT A)耗时、连线测验B(TMT B)耗时、阿尔茨海默病评估量表认知部分(ADAS-Cog)单词回忆得分高于对照组,蒙特利尔认知评估(MoCA)量表定向力、MoCA延迟回忆得分低于对照组(P<0.05)。癫痫患者脑脊液P53 mRNA相对表达水平与miRNA-34a相对表达水平呈正相关,血清P53 mRNA相对表达水平与miRNA-34a相对表达水平呈负相关(P<0.05)。癫痫患者MoCA定向力得分、MoCA延迟回忆得分与脑脊液P53 mRNA、miRNA-34a及血清P53 mRNA相对表达水平呈负相关,与血清miRNA-34a相对表达水平呈正相关(P<0.05);癫痫患者TMT A耗时、TMT B耗时、ADAS-Cog单词回忆得分与脑脊液P53 mRNA、miRNA-34a及血清P53 mRNA相对表达水平呈正相关,与血清miRNA-34a相对表达水平呈负相关(P<0.05)。结论P53 mRNA在癫痫患者脑脊液和血清中均呈高表达,miRNA-34a在脑脊液中呈高表达而在血清中呈低表达,二者均与癫痫患者认知功能相关。 Objective To explore the expression levels of cerebrospinal fluid and serum P53 mRNA and microRNA(miRNA)-34a and their correlation with cognitive function in patients with epilepsy.Methods Seventy-nine patients with epilepsy were selected as epilepsy group,and 73 patients with headache symptoms but without epilepsy were selected as control group.The expression levels of P53 mRNA and miRNA-34a in cerebrospinal fluid and serum were detected in the two groups,and the cognitive function of the patients was assessed.An analysis was performed on the correlation between P53 mRNA and miRNA-34a expression levels in cerebrospinal fluid and serum of patients with epilepsy,as well as the correlations of the relative expression levels of cerebrospinal fluid and serum P53 mRNA and miRNA-34a with cognitive function.Results The epilepsy group exhibited higher relative expression levels of P53 mRNA and miRNA-34a in cerebrospinal fluid,higher relative expression level of serum P53 mRNA,and lower relative expression level of serum miRNA-34a in contrast with the control group(all P<0.05).Compared with the control group,the epilepsy group had increased durations for Trail Making Test A(TMT A)and Trail Making Test B(TMT B),higher score for word memory domain on the Alzheimer′s Disease Assessment Scale-Cognitive Subscale(ADAS-Cog),and lower scores for orientation and delayed memory domains on the Montreal Cognitive Assessment(MoCA)scale(P<0.05).In patients with epilepsy,there was a positive correlation between the relative expression levels of P53 mRNA and miRNA-34a in cerebrospinal fluid,the relative expression level of serum P53 mRNA negatively correlated with the relative expression level of serum miRNA-34a(P<0.05).In patients with epilepsy,the scores for MoCA domains of orientation and delayed memory negatively correlated with the relative expression levels of P53 mRNA and miRNA-34a in cerebrospinal fluid and serum P53 mRNA,and positively correlated with the relative expression level of serum miRNA-34a(P<0.05);in patients with epilepsy,durations for TMT A and TMT B,score for word memory on ADAS-Cog positively correlated with the relative expression levels of P53 mRNA and miRNA-34a in cerebrospinal fluid and serum P53 mRNA,and negatively correlated with the relative expression level of serum miRNA-34a(P<0.05).Conclusion P53 mRNA is highly expressed in cerebrospinal fluid and serum of patients with epilepsy,miRNA-34a is highly expressed in cerebrospinal fluid and lowly expressed in serum,and these two indices correlate with the cognitive function of patients with epilepsy.
