摘要
目的分析不同T分期肺癌患者血清Ⅰ型T辅助细胞(Th1)/Ⅱ型T辅助细胞(Th2)平衡及基质金属蛋白酶9(MMP-9)、金属蛋白酶组织抑制剂-1(TIMP-1)水平变化情况。方法选取本院2013年1月至2018年5月收治的肺癌患者中筛选符合纳入排除标准的200例设为研究对象,应用酶联免疫吸附法检测Th1细胞干扰素-γ(IFN-γ)、Th2细胞白细胞介素-4(IL-4)水平,双抗体夹心酶联免疫吸附法检测血清MMP-9、TIMP-1水平,并计算Th1/Th2比值;比较不同T分期患者血清Th1/Th2、MMP-9、TIMP-1水平,Spearman相关性分析法分析肺癌T分期与血清Th1/Th2、MMP-9、TIMP-1的关系。结果肺癌患者血清IFN-γ、Th1/Th2随着T分期增加而下降;血清IL-4、MMP-9、TIMP-1水平随T分期上升而上升;且不同T分期及任意T分期两两比较,患者血清FN-γ、IL-4、Th1/Th2、MMP-9、TIMP-1差异均有统计学意义(P<0.05);Spearman相关性分析,Th1/Th2比值与肺癌T分期显著负相关(r=0.808,P<0.01),MMP-9、TIMP-1与肺癌T分期显著正相关(r=0.499、0.406,P<0.01)。结论血清Th1/Th2、MMP-9、TIMP-1与肺癌T分期密切相关,其或在肺癌患者肿瘤生长、扩散及浸润中发挥重要作用,值得临床重视。
Objective To analyze the balance of serum T helper cell 1(Thl)/T helper cell 2(Th2) and changes of matrix metalloproteinase-9(MMP-9) and tissue inhibitor of metalloproteinase-1(TIMP-1) levels in patients with different T stages of lung cancer.Methods From January 2013 to May 2018,200 lung cancer patients admitted to our hospital were screened as eligible for inclusion and met the exclusion criteria.The levels of Th1 cells interferon-γ(IFN-γ) and Th2 cells interleukin-4(IL-4),MMP9 and TIMP-1 were detected by enzyme-linked immunosorbent assay(ELISA).The ratio of IFN-γ to IL-4 was calculated.Serum Th1/Th2,MMP-9 and TIMP-1 levels of patients with different T stages were compared.Spearman correlation analysis was performed to analyze the relationship between T stage of lung cancer and serum Thl/Th2,MMP-9 and TIMP-1.Results Serum IFN-γ level and Th1/Th2 ratio decreased with the increase of T stage in patients with lung cancer.The levels of serum IL-4,MMP-9 and TIMP-1 increased with the increase of T stage.There were significant differences in serum FN-γ, IL-4, Th1/Th2, MMP-9 and TIMP-1 among different T stages and any two of the T stages(P<0.05).Spearman correlation analysis showed that Th1/Th2 was significantly negatively correlated with lung cancer T stage(r=-0.808,P<0.01).MMP-9 and TIMP-1 were significantly positively correlated with lung cancer T stage(r=0.499,0.406,P<0.01).Conclusion Serum Thl/Th2,MMP-9 and TIMP-1 are closely related to the T stages of lung cancer.They may involve in tumor growth,proliferation and infiltration of lung cancer patients.
