摘要
目的探讨紫杉醇(TAXOL)联合大黄素(EM)对小细胞肺癌裸鼠模型TTF-1、CD56、ProGRP的影响。方法建立小细胞肺癌裸鼠模型,根据不同的给药方式分成模型组、TAXOL组和TAXOL+EM组,每组各20只,再取20只健康鼠设空白组。检测各组裸鼠1 d、7 d、14 d的P2O2水平;HE染色检测各组肺、肝组织情况;免疫组化检测TTF-1、CD56水平,CLIA法检测ProGRP水平;RT-PCR法检测各组TTF-1、CD56.ProGRP的mRNA水平。结果持续给药可以有效提升小细胞肺癌裸鼠的PaCh水平,TAXOL+EM组效果优于TAXOL组(P<0.05);TAXOL+EM组肺、肝组织情况优于模型组和TAXOL组;TAXOL+EM组TTF-1、CD56阳性率及ProGRP水平低于模型组和TAXOL组(P<0.05),TTF-1、CD56、ProGRP的mRNA表达量也均低于模型组和TAXOL组(P<0.05)。结论大黄素可以增强紫杉醇对小细胞肺癌裸鼠TTF-1、CD56及ProGRP的影响,紫杉醇联合大黄素使用可以有效抑制小细胞肺癌。
Objective To investigate the effects of Taxol combined with emodin(EM)on TTF-1,CD56 andProGRP in nude mouse models of small cell lung cancer.Methods A nude mouse model of small cell lung cancer was established and divided into model group,Taxol group and Taxol+EM group according to difierent administration methods,with 20 mice in each group,and 20 healthy mice in each group were set as blank group.PaCh levels of nude mice in each group were detected at 1 d,7 d and 14 d.HE staining was used to detect lung and Ever tissues in each group.TTF-1 and CD56 levels were detected by immunohistochemistry,and ProGRP levels were detected by CLIA method.MRNA levels of TTF-1,CD56 and ProGRP were detected by RT-PCR.Results Continuous administration could effectively increase the level of PaO2 in nude mice with small cell lung cancer,and the eSect of Taxol+EM group was significantly better than that of Taxol group(P<0.05).The lung and liver tissues of Taxol+EM group were better than those of model group and Taxol group.The positive rates of TTF-1,CD56 and ProGRP in Taxol+EM group were significantly lower than those in model group and Taxol group(P<0.05),and the mRJSIA expression levek of TTF-1,CD56 and ProGRP in Taxol+EM group were also lower than those in model group and Taxol group(P<0.05).Conclusion Emodin can enhance the efiects of Taxol on TTF-1,CD56 and ProGRP in nude mice with small cell lung cancer,and Taxol combined with emodin can effectively inhibit small cell lung cancer.
