JAK/STAT信号通路相关蛋白在脂质肾损害大鼠肾脏组织中的表达及意义 被引量:1

Expression and significance of JAK/STAT signaling pathway-related proteins in renal tissue of rats with hyperlipidemia renal injury

摘要 目的探讨JAK/STAT信号通路相关蛋白JAK2、p-JAK2、STAT3、p-STAT3、OSM、OSMR在脂质肾损害大鼠肾脏组织中的表达及意义。方法30只SD大鼠随机分为对照组(n=15)和高脂组(n=15)。对照组给以普通饲料喂养,高脂组给以高脂饲料喂养。待喂养12周时检测两组大鼠血清甘油三脂、总胆固醇、24 h尿蛋白定量及血肌酐指标变化。ELISA方法检测两组大鼠血清中OSM浓度。光镜观察肾脏组织病理改变。免疫组化法检测JAK/STAT信号通路中OSM及其受体OSMR的表达。Western blot方法检测肾脏组织中JAK/STAT信号通路中JAK2、p-JAK2、STAT3、p-STAT3蛋白的表达。结果与对照组比较,高脂组大鼠血清甘油三酯、总胆固醇、24 h尿蛋白定量显著升高(P<0.05)。ELISA法结果显示,与对照组比较,高脂组大鼠血清中OSM大量表达(P<0.05)。与对照组比较,高脂组光镜下HE染色可见大鼠肾小球系膜细胞增多,肾小管上皮细胞浊肿;PAS染色可见肾小球系膜基质明显增多。免疫组化显示,与对照组比较,高脂组大鼠肾小管上皮细胞内OSM及其受体OSMR表达明显增多。Western blot显示,与对照组比较,高脂组大鼠肾脏组织JAK2、p-JAK2、STAT3、p-STAT3蛋白表达显著升高(P<0.05)。结论JAK/STAT信号通路相关蛋白JAK2、p-JAK2、STAT3、p-STAT3、OSM、OSMR在脂质肾损害大鼠肾脏组织中大量表达,提示JAK/STAT信号通路可能参与脂质肾损害的过程。 Objective To investigate the expression and significance of JAK/STAT signaling pathway-related proteins JAK2,p-JAK2,STAT3,p-STAT3,OSM and OSMR in renal tissue of rats with lipid renal injury.Methods Thirty SD rats were randomly divided into control group(n=15)and fatty group(n=15).The rats in control group were fed with ordinary diet,and the rats in high fat group were fed with high fat diet.After feeding for 12 weeks,serum triglyceride,total cholesterol,24 h urinary protein quantitation and serum creatinine were detected in two groups.The concentration of OSM in serum of rats was detected by ELISA in two groups.The pathological changes of renal tissue were observed under light microscope.The expression of OSM and its receptor OSMR in JAK/STAT signaling pathway was detected by immunohistochemistry.The protein expression levels of JAK2,p-JAK2,STAT3 and p-STAT3 in the JAK/STAT signaling pathway in renal tissues were detected by Western blot.Results Compared with control group,serum triglyce-ride,total cholesterol,24 h urine protein quantitation were significantly increased in high fat group(P<0.05).ELISA showed that compared with control group,OSM was highly expressed in serum of rats in high fat group(P<0.05).Compared with control group,HE staining under light microscope showed increased glomerular mesangial cells and nepheloma of renal tubular epithelial cells in high fat group.The mesangial matrix of the glomeruli was significantly increased in high fat group with PAS staining.Compared with control group,immunohistochemistry showed that the expression of OSM and its receptor OSMR in renal tubular epithelial cells was significantly increased in high fat group.Western blot showed that the protein expression levels of JAK2,p-JAK2,STAT3 and p-STAT3 in renal tissue of rats in high fat group were significantly increased compared with control group(P<0.05).Conclusion JAK/STAT signaling pathway-related proteins JAK2,p-JAK2,STAT3,p-STAT3,OSM and OSMR are highly expressed in renal tissue of rats with lipid renal injury,suggesting that JAK/STAT signaling pathway may be involved in the process of lipid renal injury.
