摘要
目的通过超高压液相色谱-三重四级杆质谱联用仪(UPLC/MS/MS)的分析方法对痛风性关节炎(gouty arthritis, GA)患者的血清进行脂质组学分析,寻找潜在的GA脂质生物标志物。方法人血清样本中加入甲醇-氯仿(体积比2∶1,含质量分数0.01%BHT)混合溶剂进行脂质提取,采用Acquity BEH C18分析柱(5 cm×1.0 mm, 1.7μm),柱温50℃,以乙腈-异丙醇(体积比为5∶2,含质量分数1%的甲酸铵)为流动相A,超纯水(含1%的甲酸铵)为流动相B,梯度方式洗脱,采用正、负离子检测电喷雾电离(±ESI)方式进行检测。对29例GA患者及22例正常人的血清样本进行脂质组学研究,比较GA患者与正常人血清中的脂质代谢物。结果 GA组和健康对照组(N组)的血清脂质指纹图谱存在明显差异;GA组与N组相比除了Lyso-GPC类的含量下降以外,其他的各类脂质均呈上升趋势,其中以GPC类和TG类最为显著;差异明显的脂类代谢物共定性鉴别出19种。结论 Lyso-GPC(18∶2)、TG(49∶3)等为代表的19种差异脂质代谢物可能为痛风性关节炎的脂类代谢标志物。
Objective To study the lipidomics characteristics of Gouty Arthritis(GA) by UPLC/MS/MS for found the lipid markers of GA.Methods The mixed solvent of methanol-chloroform(volume ratio 2∶1,0.01 % BHT) was added to human serum samples for lipid extraction, The Acquity BEH C18(5 cm×1.0 mm) was used, column temperature was 50 ℃,acetonitrile-isopropanol(volume ratio 5∶2,1 %Ammonium formate) as mobile phase A,ultrapure water(1% Ammonium formate) as mobile phase B,gradient elution, positive and negative ion mode of electrospray ionization(±ESI) was used for detection.The serum samples of 29 GA patients and 22 normal persons were studied by lipomics, and the serum lipid metabolites of patients with GA and normal persons were compared.Results Serum lipid fingerprint of GA patient and Healthy were significantly different;Compared with group N,the content of lyso-GPC in group GA showded a downward trend, but the others increased, especially GPC and TG;19 kinds of lipid metabolites with obvious difference were identified.Conclusion Lyso-GPC(18∶2),TG(49∶3) and other 19 differential lipid metabolites may be were potential lipid biomarkers of GA.
