基于网络药理学探讨当归苦参丸治疗湿疹的作用机制 被引量:2

摘要 目的利用网络药理学研究当归苦参丸治疗湿疹的作用机制。方法借助TCMSP、GeneCards、NCBI数据库,分别筛选当归、苦参的有效成分、靶点和湿疹的疾病靶点,将中药靶点与疾病靶点取交集得到共有靶点,构建蛋白质与蛋白质相互作用(protein-protein interaction,PPI)网络、中药-成分-疾病-靶点网络并分别导入cytoscape 3.8.0软件中进行拓扑分析,筛选关键靶点、关键成分,将中药-疾病共有靶点进行GO功能富集分析和KEGG通路富集分析,最后构建中药-成分-疾病-通路-靶点网络。结果筛选得到47个有效成分,83个中药-疾病共有靶点,40个关键靶点,5个关键成分;分析得到1654条生物过程,46项细胞组分,101项分子功能,123条信号通路。结论当归苦参丸可能通过影响细胞因子受体结合、细胞因子活性、受体配体活性、信号受体激活剂活性、DNA结合转录激活剂活性及RNA聚合酶Ⅱ特异性等途径发挥多成分、多靶点、多途径治疗湿疹的作用,其作用机制可能与调控糖尿病并发症中的晚期糖基化终末产物-晚期糖基化终产物受体(advanced glycation end product-receptor for advanced glycosylation end product,AGE-RAGE)信号通路、白介素(interleukin,IL)-17信号通路、肿瘤坏死因子(tumor necrosis factor,TNF)信号通路、Toll样受体(Toll-like receptor,TLR)信号通路、C型凝集素受体(C-type lectin receptor,CLR)信号通路、松弛素信号通路、T细胞受体(T cell receptor,TCR)等信号通路相关。 Objective To study the mechanism of Danggui Kushen pill in treating eczema by network pharmacology.Methods With the help of TCMSP,GeneCards and NCBI databases, the effective components and targets of Radix angelicae sinensis and Radix sophorae flavescentis, and disease targets of eczema were screened respectively.The common targets were obtained by intersection of traditional Chinese medicine(TCM) and disease targets in order to construct protein-protein interaction(PPI) and TCM-component-disease-target network which was imported into Cytoscape 3.8.0 software for topological analysis to screen key targets and components.The common targets of TCM and disease were analyzed by GO function and KEGG pathway enrichment analysis.Finally, the network of TCM-component-disease-pathway-target was constructed.Results 47 effective components, 83 common targets, 40 key targets, 5 key components, 1654 biological processes, 46 cell components, 101 molecular functions and 123 signaling pathways were obtained.Conclusion Danggui Kushen pill may play a role of multi-components, multi-targets and multi-channels in treating eczema by affecting the binding of cytokine receptor, cytokine activity, receptor ligand activity, signaling receptor activator activity, DNA binding transcription activator activity and RNA polymerase II specificity, etc.Its mechanism may be related to the regulation of advanced glycation end product-receptor for advanced glycosylation end product(AGE-RAGE) signaling pathway in diabetic complications, interleukin(IL)-17 signaling pathway, tumor necrosis factor(TNF) signaling pathway, Toll-like receptor(TLR) signaling pathway, C-type lectin receptor(CLR) signaling pathway, relaxin signaling pathway, T cell receptor(TCR) signaling pathway, etc.
