摘要 目的探讨黑色素瘤缺乏因子2(AIM2)在宫颈癌组织中的表达及机制。方法选取2017年3月至2018年5月在山东省立第三医院行宫颈癌切除术的26例患者的宫颈癌组织及癌旁组织作为研究对象,采用实时荧光定量PCR(RT-PCR)和免疫组化法检测宫颈癌及其癌旁组织中AIM2表达;采用RT-PCR和Westernblot检测人宫颈癌细胞株Hela和正常宫颈细胞株Ect1/E6E7中AIM2mRNA和蛋白表达;Hela细胞转染AIM2si-RNA(si-AIM2组)和阴性对照(si-NC组),设空白对照组和3-MA干扰组(si-AIM2+3-MA组)。Westernblot检测细胞中AIM2、LC3、Beclin1和p62的蛋白表达,CCK-8法检测细胞增殖,Transwell实验检测细胞迁移和侵袭。结果与癌旁组织比较,宫颈癌组织中AIM2的mRNA表达和IHC评分升高(P<0.001);与Ect1/E6E7组比较,Hela组中AIM2的mRNA和蛋白表达升高(P<0.001);与si-NC组比较,si-AIM2组细胞中LC3Ⅱ/LCⅠ和Beclin1蛋白表达升高(P<0.001),细胞在24h、48h和72h增殖能力(P<0.05)以及细胞迁移数和侵袭数(P<0.001)降低,AIM2和p62蛋白表达降低(P<0.001);与si-AIM2组比较,si-AIM2+3-MA组中LC3Ⅱ/LCⅠ和Beclin1蛋白表达降低(P<0.001),细胞在24h、48h和72h增殖能力(P<0.05)以及细胞迁移和侵袭数(P<0.001)升高,p62蛋白表达升高(P<0.001)。结论敲低AIM2通过诱导细胞自噬抑制宫颈癌细胞增殖、迁移和侵袭。 Objective To investigate the effect and mechanism of AIM2 on cervical cancer.Methods Cervical cancer tissues and adjacent tissues of 26 patients who underwent cervical cancer resection in Shandong Provincial Third Hospital from March 2017 to May 2018 were selected as the research objects.RT-PCR and immunohistochemistry were used to detect the expression of AIM2 in cervical cancer and its adjacent tissues;RT-PCR and Western blot were used to detect the mRNA and protein expression of AIM2 in human cervical cancer cell line Hela and normal cervical cell line Ect1/E6 E7;Hela cells were transfected with AIM2 si-RNA interference sequence(si-AIM2 group)and negative control(si-NC group),and a blank control group(blank group)and a 3-MA interference group(si-AIM2+3-MA group)were set up.Western blot was used to detect the protein expression of AIM2,LC3,Beclin1 and p62 in cells;CCK-8 was used to detect cell proliferation;Transwell experiment was used to detect cell migration and invasion.Results Compared with adjacent tissues,AIM2 mRNA expression and IHC score in cervical cancer tissues were increased(P<0.001).Compared with the Ect1/E6 E7 group,the mRNA and protein expression of AIM2 in the Hela group increased(P<0.001).Compared with the si-NC group,the expression of LC3Ⅱ/LCⅠand Beclin1 proteins in the cells of the si-AIM2 group was increased(P<0.001),and the cell proliferation capacity(P<0.05)at 24 h,48 h and 72 h,cell migration and invasion numbers(P<0.001)were decreased,the expression of AIM2 and p62 protein decreased(P<0.001);compared with the si-AIM2 group,the protein expression of LC3Ⅱ/LCⅠand Beclin1 in the si-AIM2+3-MA group decreased(P<0.001),and the cell proliferation ability(P<0.05)at 24 h,48 h and 72 h,cell migration and invasion numbers(P<0.001)were increased,and the expression of p62 protein increased(P<0.001).Conclusions Knockdown of AIM2 inhibits the proliferation,migration and invasion of cervical cancer cells by inducing autophagy.
