摘要
目的:观察针灸肺俞、大椎、风门对哮喘大鼠肺组织中嗜酸性粒细胞(EOS)凋亡及其调控蛋白Fas、Bcl-2表达的影响,从EOS凋亡角度探讨针灸治疗哮喘的作用机制。方法:将40只SD大鼠随机分为空白组、模型组、针刺组、艾灸组,每组10只。用卵蛋白对大鼠致敏、激发,建立哮喘模型。造模成功后,针刺组大鼠针刺肺俞(双)、大椎、风门(双)3穴,每次20min;艾灸组大鼠艾灸相同的穴位,每次灸10min。均每日治疗1次,连续7d,并在治疗后激发20min。模型组与空白组均行抓握刺激,不做任何治疗。标记EOS的组化染色法观察肺组织中EOS表达;免疫组化染色法观察肺组织中Fas、Bcl-2蛋白表达;TUNEL法检测肺组织中EOS凋亡。结果:与空白组比较,模型组大鼠肺组织内EOS计数显著增高,EOS凋亡率显著降低,Fas蛋白表达降低,Bcl-2蛋白表达增高(P<0.01);与模型组比较,针刺组与艾灸组大鼠肺组织内EOS计数显著降低,EOS凋亡率显著增高,Fas蛋白表达增高,Bcl-2蛋白表达降低(P<0.01)。结论:针灸可能通过提高哮喘大鼠肺组织中Fas蛋白和抑制Bcl-2蛋白表达,从而促进EOS凋亡,减轻EOS在气道内浸润,控制哮喘炎症反应。
Objective:To observe the effects of acupuncture and moxibustion of Feishu(BL 13),Dazhui(DU 14)and Fengmen(BL 12)on apoptosis of eosinophils(EOS)and the change of Fas and Bcl-2 level in lung tissue of asthmatic rats,to explore the asthma mechanism from the point of view of EOS apoptosis.Methods:Forty SD rats were randomly divided into the blank group,the model group,the acupuncture group and the moxibustion group,ten rats in each group.Sensitize and motivate with OVA to establish asthmatic model rats.After the asthmatic model was established successfully,the rats in the acupuncture group were treated with Feishu(BL 13)(double),Dazhui(DU 14)and Fengmen(BL 12)(double)for 20 minutes each time;the moxibustion group were treated with the same acupoint for 10 minutes each time.Both groups were treated once a day for 7 days,and 20 minutes was stimulated after treatment.The model group and the blank group were treated with grip stimulation without any treatment.The level of EOS in lung tissue was observed by histochemical staining;the level of Fas and Bcl-2 protein in lung tissue was observed by immunohistochemical staining;and the apoptosis of EOS in lung tissue was observed by TUNEL.Results:Compared with the blank group,the EOS count in the lung tissue of the model group was significantly increased,the apoptosis rate of EOS was significantly decreased,the Fas protein was decreased,and the Bcl-2 protein was increased(P<0.01).Compared with the model group,the EOS count in the lung tissue of the acupuncture and the moxibustion group was obviously reduced,the apoptosis rate of the EOS was obviously increased,the Fas protein was increased,and the Bcl-2 protein was reduced(P<0.01).Conclusion:Acupuncture and moxibustion may increase the level of Fas protein and inhibit the level of Bcl-2 protein in lung tissue of asthma model rats so as to promote apoptosis of EOS,reduce EOS infiltration in airway and control inflammatory reaction of asthma.
