摘要
目的:研究芦荟与火麻仁配伍对复方地芬诺酯致便秘小鼠的通便作用。方法:将SPF级健康雄性昆明小鼠随机分为8组:空白对照组,模型组,阳性对照组,芦荟烘干粉组,火麻仁油组,芦荟烘干粉+火麻仁油低、中、高剂量组(配伍组低、中、高剂量组,配伍剂量比均为0.9∶2)。各组预防给药灌胃相应药物,1次/d,连续14d。取材前1h,除空白对照组外,其余各组灌胃复方地芬诺酯制备便秘模型。观察小鼠一般状况,排便情况包括首粒排黑便时间、5h内排便粒数及重量,ELISA法检测结肠组织5-羟色胺(5-HT)、P物质(SP)、血管活性肠多肽(VIP)含量变化;RT-PCR检测结肠组织水通道蛋白(AQP)3、AQP9 mRNA表达水平;Western Blot检测AQP3、AQP9蛋白表达水平。结果:与模型组比较,各给药均能显著缩短小鼠首粒排黑便时间(P<0.01),增加5h内黑便粒数及黑便重量(P<0.01);显著升高结肠组织内5-HT、SP含量(除火麻仁油组5-HT外)(P<0.01,P<0.05),降低V IP含量(P<0.01);配伍组中剂量组能显著上调AQ P9 m R NA表达以及蛋白水平(P<0.05,P<0.01),下调AQP3 mRNA表达以及蛋白水平(P<0.05,P<0.01)。结论:芦荟与火麻仁配伍可显著改善复方地芬诺酯造成的小鼠便秘情况,缩短粪便在肠道内停留时间,两者配伍可能通过调节肠道神经递质5-HT、VIP、S P含量,增强胃肠道动力;降低AQ P3、升高AQ P9表达,减少肠道对水的重吸收,增加肠液分泌,共同达到通便效果。
Objective:To study the laxative effect of Aloe combination with Cannabis Fructus on compound diphenoxylate-induced constipation in mice.Methods:SPF-grade healthy male Kunming mice were randomly divided into 8 groups:blank control group,model group,positive control group,Aloe drying powder group,Cannabis Fructus oil group,and Aloe drying powder+Cannabis Fructus oil low,medium and high dose group(compounded group low,medium and high dose group),and the compounded dose ratio was 0.9∶2).Each group was administered prophylactically by gavage of the corresponding drug once a day for 14d.One hour before sampling,the constipation model was prepared by gavage of compound diphenoxylate in all groups except the blank control group.The general conditions of mice were observed,including the time of the first black stool,the number and weight of pellets defecated within 5h.The changes of 5-HT,SP and VIP contents in colonic tissues were detected by ELISA;the mRNA expression levels of aquaporin AQP3 and AQP9 in colonic tissues were detected by RT-PCR;the protein expression levels of both were detected by Western Blot.Results:Compared with the model group,each administration group could significantly shorten the first black stool time(P<0.01),increased the number and weight of black stool within 5h(P<0.01);significantly increased the 5-HT and SP content in colonic tissue(except 5-HT in Cannabis Fructus oil group)(P<0.01,P<0.05)and decreased the VIP content(P<0.01).The AQP9 mRNA expression and protein level were significantly up-regulated(P<0.05,P<0.01)and AQP3 mRNA expression and protein level were down-regulated(P<0.05,P<0.01).Conclusion:The combination of Aloe and Cannabis Fructus significantly improve the constipation of mice caused by compound diphenoxylate and shorten the residence time of feces in the intestine.The combination of the two may enhance the gastrointestinal motility by regulating the content of intestinal neurotransmitters 5-HT,VIP and SP;reduce the expression of aquaporin AQP3 and increase the expression of AQP9,reduce the reabsorption of water in the intestine and increase the secretion of intestinal fluid,which together achieve the laxative effect.
