益气清热膏对氨基核苷嘌呤霉素肾病综合征大鼠模型肾组织Wnt/β-catenin信号通路表达的影响 被引量:1

Effects of Yiqi Qingre Cream on Wnt/β-catenin signaling pathway in renal tissues of puromycin aminonucleoside nephrosis rat model

摘要 目的:探讨益气清热膏对氨基嘌呤霉素肾病综合征模型Wnt/β-catenin信号通路相关分子表达的影响。方法:采用RT-PCR法及Western Blot法检测PAN模型造模后第3、10、15天大鼠肾组织中wnt4、wnt5a、β-catenin、GSK-3β、FEN的基因和蛋白表达情况。结果:造模后第15天,与模型组比较,益气清热膏可以抑制PAN大鼠模型wnt4、β-catenin、GSK-3β和FEN蛋白的表达(P<0.05,P<0.01),并且随着治疗时间延长,益气清热膏作用靶点逐渐增多。结论:益气清热膏可以降低PAN大鼠模型肾组织wnt4、β-catenin、GSK-3β、FEN的基因和蛋白表达,这可能是其降低蛋白尿、修复足细胞损伤的作用机制之一。 Objective:To explore the effects of Yiqi Qingre Cream by investigating alterations of the molecule of Wnt/β-catenin signaling pathway in puromycin aminonucleoside nephrosis(PAN)rat model.Methods:RT-PCR and Western Blot analysis were conducted to detect the expressions of wnt4,wnt5a,β-catenin,GSK-3β,FEN of renal tissues were detected on the 3rd,10th and 15th day after PAN modeling.Results:Compared with model group,the protein expression levels of wnt4,β-catenin,GSK-3βand FEN was inhibited by Yiqi Qingre Cream(P<0.05,P<0.01)with the extension of treatment time,the target of Yiqi Qingre Cream gredually increased.Conclusion:Yiqi Qingre Cream can inhibit wnt4,β-catenin,GSK-3β,and FEN expression in PAN model,which may be related with its therapeutic mechanisms of reducing proteinuria and alleviating podocyte damage.
