探讨甲基化芯片技术检测粪便DNA甲基化用于大肠癌早期筛查的作用 被引量:2

摘要 目的 探讨甲基化芯片技术检测粪便DNA甲基化用于大肠癌早期筛查的作用。方法 选取本院2018年6月-2019年11月确诊的60例大肠癌患者为观察组,另外选取同期于本院进行体检的健康受试者60例为对照组。结果 OD260/280值在1. 8~2. 0以及凝胶电泳图结果表明,粪便中提取的DNA可用于研究对比。观察组患者粪便Vimentin、s FRP1、h MLH1基因甲基化水平及甲基化阳性水平均显著高于对照组(P <0. 05),甲基化阴性水平显著低于对照组(P <0. 05)。采用甲基化芯片检测粪便h MLH1基因甲基化法检测对诊断大肠癌的敏感度为83. 33%,明显高于MSP法、FOBT和CEA,上述差异均有统计学意义(P <0. 05)。结论 甲基化芯片技术检测粪便DNA甲基化对于大肠癌早期筛查的检出率较高,优于临床上的MSP法、FOBT和CEA法,可作为临床上大肠癌早期筛查的良好手段进行推广。 Objective To investigate the role of DNA methylation chip technology in early screening of colorectal cancer.Methods Sixty patients with colorectal cancer diagnosed in our hospital from June 2018 to November 2019 were selected as the observation group,and 60 healthy subjects for physical examination in our hospital during the same period were selected as the control group.Results The results of this study indicated that the OD_(260/280) value ranged from 1.8 to 2.0,and the results of gel electrophoresis showed that the DNA extracted from feces could be used for study and comparison.The methylation level and positive level of Vimentin,sFRP1 and hMLH1 genes in feces of the observation group were significantly higher than that of the control group(P<0.05),and the negative level of methylation was significantly lower than that of the control group(P<0.05).The sensitivity of detection of fecal hMLH1 gene methylation by methylation chip was 83.33%,which was significantly higher than that of MSP method,FOBT and CEA,with the above differences statistically significant(P<0.05).Conclusion The detection rate of fecal DNA methylation by the methylation chip technique for early screening of colorectal cancer is relatively high,which is superior to the clinical MSP method,FOBT method and CEA method,and can be promoted as a good clinical method for early screening of colorectal cancer.
