运脾泻肺化痰汤对幼龄哮喘大鼠气道黏液高分泌的影响 被引量:6

Effect of Yunpi Xiefei Huatan Decoction on airway mucus hypersecretion in young asthmatic rats

摘要 目的探讨运脾泻肺化痰汤对幼龄哮喘大鼠气道黏液高分泌的调节与促进气道核因子κB(NF-κB)信号通路信的核因子κB抑制蛋白α(IκBα)分泌和黏蛋白5AC(MUC5AC)表达的影响。方法用随机数字表法分为4组:正常组、模型组、中药组(运脾泻肺化痰汤)和西药组(布地奈德),每组10只。除正常组外,其他3组通过卵清白蛋白联合氢氧化铝致敏雾化激发建立幼龄哮喘大鼠模型,于1天和第8天致敏,第15天开始雾化激发隔天1次,共激发7次。第16天在造模的同时,正常组、模型组均灌胃0.9%NaCl 2 mL,中药组灌胃运脾泻肺化痰汤20μL·g^(-1)灌胃,西药组灌胃予0.01%布地奈德雾化吸入20 min,连续4周。以逆转录-PCR法检测大鼠肺组织中的IκBα、P65和MUC5AC mRNA表达量。结果正常组、模型组、中药组和布地奈德组的IκBα基因的表达量分别为1.12±0.14,0.27±0.05,0.66±0.11和0.52±0.09;这4组的P65基因的表达量分别为1.12±0.27,4.82±0.76,2.82±0.59和2.85±0.90;这4组MUC5AC基因的表达量分别为1.12±0.32,5.84±0.66,3.28±1.06,4.35±0.65。模型组与正常组比较,上述基因的表达差异均有统计学意义(均P<0.05),中药组与西药组比较,上述基因的差异均有统计学意义(均P<0.05)。结论运脾泻肺化痰汤有效减轻哮喘气道炎症其治疗机制之一可能是通过促进IκB mRNA表达,抑制核因子-κB信号通路激活,降低MUC5AC mRNA的相关表达量。 Objective To investigate the effect of Yunpi Xiefei Huatan Decoction(YXHD)on airway mucus hypersecretion in young asthmatic rats,the correlation between the promotion of airway nuclear factorκB(NF-κB)signaling pathway inhibitor of nuclear factor kappa Bα(IκBα)secretion and the expression of mucin 5AC(MUC5AC).Methods According to random number table,SD rats were randomly divided into 4 groups:normal group,model group,Chinese herbal medicine(CHM)group,western medicine group,with 10 rats in each group.Except the normal group,the other groups were sensitized by ovalbumin(OVA)combined with alumina hydroxide to establish the young asthma rat model,which was sensitized on the 1st and 8 th day.On the 15 th day,the atomized stimulation was started once every other day,and a total of 7 times were stimulated.On day 16,at the same time of modeling,normal group and model group were gavaged with 0.9%Na Cl 2 m L,CHM group were gavaged with YXHD20μL·g^(-1),western medicine group were gavaged with 0.01%budesonide atomization inhalation for 20 min,for 4weeks.The mRNA expression of IκBα,P65 and MUC5AC were detected by reverse transcriptase-PCR.Results The expression of IκBαgene in normal group,model group,CHM group and western medicine group were1.12±0.14,0.27±0.05,0.66±0.11 and 0.52±0.09,respectively;the expression of P65 gene in the four groups were 1.12±0.27,4.82±0.76,2.82±0.59 and 2.85±0.90 respectively;the expression of MUC5AC gene in the four groups were 1.12±0.32,5.84±0.66,3.28±1.06 and 4.35±0.65,respectively.The above indicators:there were statistically significant differences between the model group and the normal group(all P<0.05),and there were statistically significant differences between CHM group,western medicine group and model group(P<0.05,P<0.01).Conclusion YXHD can effectively reduce the airway inflammation of asthma,one of its therapeutic mechanisms may be through promoting IκB mRNA expression,inhibition of nuclear factor-κB signaling pathway was activated to reduce the expression of MUC5AC mRNA.
