干扰WNT5B抑制胃癌细胞的集落形成、侵袭和迁移

摘要 目的探索无翅型MMTV整合位点家族成员5B(wingless-type MMTV integration site family member 5B,WNT5B)对胃癌细胞的集落形成、侵袭和迁移的影响及其机制。方法采用蛋白质免疫印迹法检测WNT5B在癌旁非肿瘤胃组织和胃肿瘤组织、人正常胃粘膜细胞(GES-1)以及胃癌细胞(MGC-803、SGC-790、AGS和MKN-45)中的表达情况;采用shRNA法干扰SGC-790和AGS细胞中WNT5B的表达后,分别采用集落形成法、划痕法和Transwell小室法检测胃癌细胞的集落形成率、迁移和侵袭;采用免疫荧光法和蛋白质免疫印迹法检测β-catenin信号通路相关蛋白的表达水平。结果WNT5B在胃肿瘤组织和胃癌细胞中高表达。干扰WNT5B可抑制胃癌细胞的集落形成、迁移和侵袭并下调β-catenin、c-Myc和cyclinD1表达水平。结论干扰WNT5B可能通过降低β-catenin信号通路活性抑制胃癌细胞的集落形成、侵袭和迁移。 Objective To explore the role of wingless-type MMTV integration site family member 5B(WNT5B)in colony formation,invasion and migration of gastric cancer cells and the relevant mechanism.Methods The expression of WNT5B in adjacent non-tumor gastric tissues,gastric tumor tissues,human normal gastric mucosa cells(GES-1)and gastric cancer cells(MGC-803,SGC-790,AGS and MKN-45)was detected by Western blot.The expression of WNT5B in SGC-790 cells and AGS cells was silenced by the shRNA method,and the colony formation rate,migration and invasion of GC cells were detected by colony formation assay,scratch assay and Transwell assay,respectively.The expression level ofβ-catenin signaling pathway-related proteins was detected by immunofluorescence assay and Western blot.Results WNT5B was highly expressed in gastric cancer tissues and cell lines.Interference with WNT5B inhibited colony formation,migration and invasion of gastric cancer cells and downregulated the expression level ofβ-catenin,c-Myc and cyclinD1.Conclusion Interference with WNT5B inhibits colony formation,invasion,and migration of gastric cancer cells by reducing the activity ofβ-catenin signaling pathway.
