高效氯氟氰菊酯聚多巴胺微胶囊悬浮剂对中华绒螯蟹非特异性免疫相关基因表达及肠道菌群的影响 被引量:1

摘要 纳米科技近些年来发展迅速,利用纳米技术合成的纳米农药具有广泛的应用前景,然而纳米农药对水生生物影响的相关报道却很有限.本文以中华绒螯蟹(Eriocheir sinensis)为研究对象,探讨不同浓度(0,0.012,0.06,0.12,0.60 mg/L)高效氯氟氰菊酯聚多巴胺微胶囊悬浮剂(LC/PDA-MS)及空白PDA悬浮剂在暴露7 d、14 d、21 d后,对中华绒螯蟹肝胰腺与血细胞中免疫相关基因表达量的影响以及21 d实验处理后LC/PDA-MS对肠道微生物群落组成的影响.研究结果显示:实验组中肝胰腺和血细胞中前酚氧化酶原(Propo)、溶菌酶(LZM)、髓样分化因子(myD88)、酚氧化酶激活因子(PPAF)、含半胱氨酸的天冬氨酸水解酶(Caspase)、Crustin这几个蛋白对应基因的表达量随着时间的增加整体呈现下降的趋势;空白悬浮剂对免疫相关基因影响较小;肝胰腺中免疫基因的表达量较血细胞的表达量高.另外,在针对实验组中华绒螯蟹肠道菌群的分析中,拟杆菌门(Bacteroidetes)在0.60 mg/L和空白悬浮剂组中丰度高;基于属水平的分析,希瓦氏菌属(Shewanella)、气单孢菌属(Amromonas)的含量降低.以上结果表明:LC/PDA-MS可以改变肝胰腺和血细胞中免疫相关基因的表达量并对肠道菌群的平衡有一定的影响. Nanotechnology is an emerging area of science that involves the engineering of nanosize particles of various materials which has a potential to play a major role in green agriculture.The nanopesticides composed of organic molecular active ingredient nanocarrier complexes have been widely applied in agriculture.However,the effects of nanopesticide on aquatic animals is rarely reported.In present study,Chinese mitten crabs Eriocheir sinensis were raised under exposure to varied concentrations(0,0.012,0.06,0.12,0.60 mg/L)of lambda-cyhalothrin polydopamine microcapsule suspension(LC-PDA)for 7,14,21 days to explore the effects of LC-PDA on related immune gene expression and intestinal microflora composition.The blank suspension was taken as a control.The results showed a downward trend for prophenoloxidase(Propo),lysozyme(LZM),myD88,PPAF,caspase,crustin gene expression along the increase of exposure time.There was little side effects under the blank LC-PDA suspension exposure.Besides,the immune gene expression in hepatopancreas was higher than that in haemocytes.We further studies the relative abundance of Bacteroidetes in 0.60 mg/L and blank PDA suspension,the results showed a decrease in Shewanella and Amromonas genes in comparison with that in control.It can be concluded that LC-PDA microcapsule suspension influences the expression of immune gene both in hepatopancreas and haemocytes.The composition of microflora in intestine is related with LC-PDA.
