红核妇洁洗液对大鼠胚胎-胎仔发育毒性研究

Evaluation of the toxicity of Honghe Fujie Lotion for embryo-fetal development

目的探讨经阴道冲洗途径给予红核妇洁洗液对大鼠胚胎-胎仔的发育毒性作用。方法2020年10月至2021年3月在西安交通大学医学院完成实验。SD孕鼠随机分为4组:红核妇洁洗液高、中、低3个剂量实验组和对照组,每组20~21只。各实验组孕鼠于受孕第6至第15天经阴道冲洗途径按1.0 mL/kg体重剂量给予红核妇洁洗液原液、2倍稀释液和4倍稀释液,2次/d;对照组孕鼠给予同样容积的生理盐水。各组孕鼠于妊娠第0、5~15、20天记录体重;妊娠第20天,颈椎脱臼处死,计数活胎数、死胎数和吸收胎数;观察胎仔外观、称量其体重后,将每窝1/2的活胎仔乙醇固定、茜素红染色、甘油透明后检查骨骼发育情况;另1/2活胎仔经Bouins液固定后,检查内脏发育情况。结果在实验剂量范围内,红核妇洁洗液各剂量组孕鼠的生殖能力、胚胎形成和胎仔外观、骨骼及内脏生长发育与对照组相比差异均无统计学意义(P>0.05)。结论在上述实验条件下,未见红核妇洁洗液阴道冲洗对大鼠胚胎-胎仔的发育毒性作用。

Objective To evaluate the maternal and fetal toxicity of Honghe Fujie Lotion in SD rats.Methods SD pregnant rats were randomly divided into 4 groups:high-,medium-,or low-dosage Honghe Fujie Lotion groups and control group,20-21 rats in each group.Pregnant rats in each experimental group were given Honghe Fujie Lotion stock solution,2-fold diluent and 4-fold diluent by vaginal flushing at the dose of 1.0 ml/kg from the 6 th to 15 th day of pregnancy,twice a day;the pregnant rats in the control group were given the same dose of normal saline.The weight of pregnant rats was recorded at d0,d5-d15 and d20.The number of live fetus,the number of dead fetus and the number of absorbed fetus were counted at d20 after exection.After observing the appearance of the fetus and weighing the weight of the fetus,the skeletal development was examined after 1/2 live fetus of each litter was fixed with alcohol,alizarin red stain and glycerine transparency.The other 1/2 live fetuses were fixed with Bouins solution and the development of viscera was examined.Results In the range of experimental dose,there were no significant differences in reproductive ability,embryo formation,fetal appearance,or bone and viscera growth and development between the groups of Honghe Fujie Lotion and the control group(P>0.05).Conclusion There is no toxicity of Honghe Fujie Lotion(2,1,0.5 mL/kg)to rat maternal body and embryo and fetal development.
