血虚风燥型特应性皮炎患者血清差异表达miRNA生物信息学筛选 被引量:3

Bioinformatics screening of differentially expressed serum miRNA in atopic dermatitis patients with syndrome of blood deficiency and wind-dryness

摘要 目的:筛选血虚风燥型特应性皮炎(AD)患者与健康人血清差异表达的miRNA,并预测其调控的靶基因和功能。方法:纳入6例血虚风燥型AD患者和6例健康志愿者,利用高通量测序技术检测两组患者血清miRNA的相对表达量,筛选差异表达miRNA,并利用生物信息学分析对差异最大的6个miRNA进行靶基因预测和功能富集分析。结果:筛选出血虚风燥型AD患者差异表达miRNA共74个,其中表达上调34个,表达下调40个。在线数据库预测公共基因1045个,GO分析显示这些miRNA的靶基因主要参与调控基因的表达与转录及蛋白质的结合等;KEGG通路富集分析显示预测靶基因参与了TGF-β、Hippo、PI3K-Akt等信号通路。结论:血虚风燥型AD患者血清中存在特异性表达的miRNA。 Aim:To predict and analyze target genes and functions by screening the differentially expressed miRNA in serum between healthy people and atopic dermatitis(AD)patients with syndrome of blood deficiency and wind-dryness.Methods:High throughput sequencing technology was used to detect the relative expression levels of miRNA in serum of 6 AD patients with blood deficiency and wind-dryness and 6 healthy volunteers,and the differentially expressed miRNA was screened.Target gene prediction and functional enrichment analysis were performed for the 6 miRNA with the highest differential multiple.Results:A total of 74 differentially expressed miRNA were screened out in AD patients with syndrome of blood deficiency and wind-dryness,among which,34 upregulated and 40 downregulated.The online databases predicted 1045 public genes,and GO enrichment showed that these miRNA target genes were mainly involved in regulating gene expression and transcription,protein binding,etc.KEGG pathway enrichment showed that the predicted target genes were involved in TGF-β,Hippo,PI3K-Akt and other signaling pathways.Conclusion:Specific miRNA expression exists in serum from AD patients with syndrome of blood deficiency and wind-dryness.
