Bioinformatics study on potential mechanisms for the development of diabetic nephropathy to end-stage renal disease

摘要 目的利用生物信息学方法探讨糖尿病肾病发展为终末期肾病的差异基因表达和潜在机制。方法检索获取美国国家生物技术信息中心基因表达综合数据库并获取糖尿病肾病和终末期肾病外周血单核细胞的基因表达芯片GSE142153数据集,筛选样本差异表达基因数据进行富集分析并筛选核心靶点,利用基因集富集分析方法筛选糖尿病肾病和终末期肾病差异表达基因富集的生物学功能。结果糖尿病肾病发展为终末期肾病的过程中,差异基因以上调表达为主,显著富集于氮素代谢信号通路;涉及蛋白质-谷氨酰胺γ-谷氨酰转移酶活性等分子功能、血红蛋白复合物等细胞组分和红细胞分化等生物学过程。结论糖尿病肾病发展为终末期肾病差异基因异基因富集于氮素代谢信号通路;涉及蛋白质-谷氨酰胺γ-谷氨酰转移酶活性等分子功能、血红蛋白复合物等细胞组分和红细胞分化等生物学过程。 Objective To explore the differential gene expression and potential mechanisms for the development of diabetic nephropathy to end-stage renal disease by the bioinformatics method.Methods The gene expression comprehensive database of National Center for Biotechnology Information was searched and the gene expression microarray GSE142153 dataset of peripheral blood mononuclear cells from diabetic nephropathy and end-stage renal disease was obtained.The data of differentially expressed genes in sample were screened for enrichment analysis and the core targets were screened.The biological functions of differentially expressed gene enrichment in diabetic nephropathy and end-stage renal disease were screened by gene set enrichment analysis method.Results When diabetic nephropathy developed to end-stage renal disease,differential genes were predominantly up-regulated and significantly enriched in nitrogen metabolic signaling pathways.This development involved molecular functions such as protein-glutamineγ-glutamyl transferase activity,cellular components such as hemoglobin complex,and biological processes such as erythrocyte differentiation.Conclusion When diabetic nephr opathy develops to end-stage renal dis ease,the differ ential genes are enriched in nitrogen metabolic signaling pathways.This development involves molecular functions such as proteinglutamineγ-glutamyl transferase activity,cellular components such as hemoglobin complex and biological processes such as erythrocyte differentiation.
