鼠尾胶原的高效提取与纯化技术

摘要 I型胶原蛋白是细胞生物学中广泛应用的细胞外基质蛋白之一,也是组织工程等领域常用的生物活性材料,是生物学、细胞力学实验室大量使用且必需的蛋白试剂,一般从大鼠鼠尾中提取获得,称为鼠尾胶原。通过抽取鼠尾肌腱、醋酸溶解、蛋白透析纯化等操作,建立了一种鼠尾胶原的高效提取与纯化技术,能简便、快速地分离得到高纯度的鼠尾胶原。经检测,利用该方法得到的鼠尾胶原具有与商用鼠尾胶原极其相似的实际使用效果。该方法可节约大量实验经费,具备推广价值,同时涉及多种基础实验操作,亦可作为实验教学内容。 Collagen I is one of the extracellular matrix proteins widely used in cell biology as well as a bioactive material commonly applied in tissue engineering, which is generally extracted from rat tails. Collagen I is an essential protein reagent existing in biology and cell mechanics laboratories. An efficient, rapid and easy operation extracting and purifying technology collagen I from rat tail has been designed and developed. The high-purity collagen is extracted by separation of rat tail tendon and dissolution in acetic acid, and then purified by protein dialysis. Feasibility validation experiments show that protein prepared by this method has similar practical effect as commercial collagen. This method saves experimental expenditure and has popularizing value. It also involves a variety of basic experimental operation skills and is suitable for experimental teaching project.
