黄连解毒汤中盐酸小檗碱、黄芩苷、黄柏碱栀子苷的含量测定

Determination of berberine hydrochloride,baicalin,ephedrine and gardeniside in huanglian detoxification soup

目的测定黄连解毒汤中盐酸小檗碱黄芩苷、黄柏碱、栀子苷主要化学成分含量,为经典名方-黄连解毒汤开发及质量控制研究提供参考。方法采用高效液相色谱法进行含量测定,盐酸小檗碱以乙腈-0.05mol/L磷酸二氢钾(25:75)为流动相,检测波长为345nm,柱温35℃;黄芩苷以甲醇-0.2%磷酸水(47:53)为流动相,检测波长为280nm,柱温35C;黄柏碱以乙腈-0.1%磷酸水(13:87)为流动相,检测波长为284nm柱温35℃;梔子苷以乙腈-水(13:87)为流动相,检测波长为238nm,柱温35℃。结果盐酸小檗碱在0.09~0.93μg,黄芩苷在0.15~1.85μg、黄柏碱在0.24~1.91μg、栀子苷在0.09~1.13μg范围内呈良好的线性关系,回收率分别为100.37%(RSD=2.43%,n=6)、99.06%(RSD=1.41%,n=6)、99.94%(RSD=1.86%,n=6)、9940%(RSD=2.28%,n=6),精密度稳定性、重现性均符合要求。结论各方法可以用于黄连解毒汤中的盐酸小檗碱、黄芩苷、黄柏碱栀子苷的含量测定。

Objective The main chemical content of berberine hydrochloride, baicalin, flavalin and gardeniside was determined in huanglian detoxification soup, providing a reference for the development and quality control research of classic prescription-huanglian detoxification soup. Methods Content was determined by HPLC. Berberine hydrochloride took potassium acetonitrile-0.05 mol/L dihydrogen phosphate(25∶75)as the flow phase of 345 nm, column temperature of 35℃, baicalin with methanol-0.2% phosphate water(47 ∶53), column temperature of 35℃, and acetonitrile-0.1% phosphate water(13 ∶87), the detection wavelength of 284 nm column temperature of 35℃, and acetonitrile-water(13∶87) as the flow phase of238 nm, column temperature of 35℃. Results Berberine hydrochloride was well linear in 0.09~0.93 μg,baicalealin in 0.15~1.85μg, baicalin in 0.24~1.91μg, gardeniine in 0.09~1.13μg, with recovery rate of100.37%(RSD=2.43%, n=6), 99.06%(RSD=1.41%, n=6), 99.94%(RSD= 1,86%, n=6), 9940%(RSD=2.28%,n=6), precision, stability and reproducibility met the requirements. Conclusion Each method can be used for the content determination of berberine hydrochloride, baicalin, ephedrine and gardeniside in Huanglian detoxification soup.
