有氧糖酵解和上皮-间充质转化在肿瘤转移机制中的作用 被引量:5

The role of glycolysis and epithelial-mesenchymal transition in tumor metastasis

摘要 有氧糖酵解是恶性肿瘤的特征之一,该能量代谢方式的改变可缩短供能周期,提高供能速率,而葡萄糖不完全氧化导致的中间产物富余又能使肿瘤细胞从中获益。上皮-间充质转化(EMT)则作为介导肿瘤侵袭转移的初始步骤,期间常伴有各种标志蛋白和转录因子的改变。此外,有氧糖酵解除了为肿瘤细胞供给能量,还可以通过该过程中的限速酶和创造肿瘤微环境来诱导EMT,促进肿瘤细胞侵袭转移,而EMT相关转录因子又可以正性调节有氧糖酵解通量,两者互作互利。本文旨在阐述有氧糖酵解和EMT在肿瘤转移机制中的作用,进而深入理解肿瘤转移机制的多态性并探索更精准的治疗靶点。 Aerobic glycolysis is one of the characteristics in malignant tumor.the change of energy metabolism can make the energy supply cycleshorten as well as the velocity up,while the surplus of intermediates caused by incomplete oxidation of glucose can benefit tumor cells.Epithelial-mesenchymal transition(EMT)as the initial step of mediating tumor invasion and metastasis,is often accompanied by changes in various marker proteins and transcription factors.Therefore,glycolysis can not only supply energy to tumor cells,but also accelerate invasion and metastasis through inducing EMT by creating tumor microenvironment and rate-limiting enzymes in this process.Moreover,EMT-related transcription factors(EMT-TFs)can promote the flux of aerobic glycolysis,they interact with each other for mutual benefit.This review focus on the of glycolysis and epithelial-mesenchymal transition in the mechanism of tumor metastasis,then deeply realize the polymorphism of the metastasis in malignancies and explore the therapeutic targets more exactly.
