摘要
目的探讨APN基因3’非编码区(3’UTR)变异与中国人群T2DM的相关性。方法对50名非DM人群(NDM)进行APN基因3’UTR测序。根据测序结果,对T2DM患者730例(T2DM组)和NDM人群612名(NDM组)的APN基因3’UTR 8个变异位点(rs56354395、rs6773957、rs1063537、rs2082940、rs1063538、rs4686804、rs1063539和rs17846865)进行基因分型和比较。结果两组APN基因3’UTR rs56354395、rs6773957、rs1063537、rs2082940、rs1063538、rs4686804、rs1063539和rs17846865基因型频率和等位基因频率经Bonferroni校正后,差异无统计学意义(P>0.006)。NDM组中,与rs17846865位点C/C+C/T基因型相比,T/T基因型具有更低的FPG[(4.84±0.56)vs(5.03±0.51)mmol/L,P<0.01]和Hb A1c[(5.10±0.39)%vs(5.21±0.35)%,P=0.01)],更高的LDL-C[(2.23±0.71)vs(2.06±0.55)mmol/L,P=0.01]。结论 APN基因3’UTR变异与中国人群T2DM无相关性,rs17846865位点可能与中国人群糖脂代谢指标相关。
Objective To investigate the association between adiponectin(APN)gene 3’untranslated region(3’UTR)variations and type 2 diabetes mellitus(T2DM)in Chinese population.Methods Firstly,the adiponectin gene 3’UTR were sequenced in 50 non diabetes mellitus(NDM)subjects.Then,according to the sequencing results,eight variations of adiponectin gene 3’UTR(rs56354395,rs6773957,rs1063537,rs2082940,rs1063538,rs4686804,rs1063539,rs17846865 and rs6414520)were selected and genotyped in T2DM subjects(n=730)and NDM subjects(n=612).Results For the eight variations,the distribution of alleles and genotypes were no differences between the T2DM and NDM groups(P>0.006).In the NDM group,for the rs17846865,compared with the C/C+C/T genotype,T/T genotype individuals showed a lower FPG[(4.84±0.56)vs(5.03±0.51)mmol/L,P<0.01]and Hb A1c[(5.10±0.39)vs(5.21±0.35)%,P=0.01],and higher LDL-C[(2.23±0.71)vs(2.06±0.55)mmol/L,P=0.01].Conclusion The 3’UTR variations in APN gene were not associated with T2DM,but rs17846865 could be associated with the glucose and lipid metabolism in Chinese population.
