摘要 目的建立同时测定三角草中异荭草苷和异牡荆素成分的高效液相色谱方法,为三角草质量评价提供参考。方法采用岛津高效液相色谱仪Shimadzu SPD-20AT,色谱柱:Agilent Ecilipse XDB-C_(18)柱(4.6 mm×250 mm,5.0μm);柱温为35℃,检测波长:339 nm,流动相为0.1%甲酸(A)-乙腈(B),梯度洗脱:7%~15%B(0~8 min),15%~20%B(8~12 min),20%~26%B(12~20 min),26%~35%(20~28 min),35%~50%B(28-35 min),50%~7%B(35~50 min),流速:1.0 mL·min^(-1)。结果异荭草苷和异牡荆素分别在0.049~0.738μg和0.044~0.660μg线性关系良好,加样回收了分别为100.70%和100.38%,两者的RSD分别为1.74%和1.83%。结论本研究建立的HPLC同时测定三角草中异荭草苷和异牡荆素成分含量方法,操作简单、结果准确、重复性好,可用于三角草的质量控制。 OBJECTIVE To establish an HPLC method for the simultaneous determination of contents of isoorientin and isovitexin in Chlorophytum laxum R.Br,and to provide references for quality evaluation of Chlorophytum laxum R.Br.METHODS Agilent Ecilipse XDB-C_(18) column(250 mm×4.6 mm,4μm)was used,with 0.1%formic acid(A)-acetonitrile(B),gradient elution〔7%-15%B(0-8 min),15%-20%B(8-12 min),20%-26%B(12-20 min),26%-35%B(20-28 min),35%-50%B(28-35 min),50%-7%B(35-50 min)〕as the mobile phase;the flow rate was 1.0 mL·min^(-1);the column temperature was maintained at 35℃;the detection wavelength was set at 339 nm;the injection volume was 20μL.RESULTS Isoorientin and isovitexin were in good linear relationship in the range of 0.0049-0.738μg and 0.0044-0.660μg respectively.Sample recovery rates were 100.70%and 100.38%;and RSDs were 1.74%and 1.83%respectively.CONCLUSION The established HPLC method for the simultaneous determination of isoorientin and isovitexin in Chlorophytum laxum R.Br is simple,accurate,and repeatable,and can be used for quality control of Chlorophytum laxum R.Br.
