摘要
为了解鸡呼吸道相关性淋巴组织(RALT)免疫功能的建立情况并反映鸡在此阶段呼吸道的免疫状态,本试验通过实时荧光定量PCR方法,检测鸡在胚胎时期和出壳后初期RALT中IL-2基因和IFN-γ基因mRNA的表达水平。结果显示,胚胎时期以及出壳后1,4日龄时,RALT中细胞因子IL-2和IFN-γmRNA的表达水平极低。随着日龄的增长,IL-2和IFN-γmRNA表达水平逐渐增加,并且在14日龄时迅速增加至较高水平,并在21日龄时基本达到峰值。各时期同一RALT中IFN-γ的表达水平都远高于IL-2。此外,对同时期喉、气管和肺脏3种器官中IL-2和IFN-γmRNA表达水平的比较发现,肺脏的表达水平远高于其他器官。结果表明,随着生长发育RALT的细胞免疫功能逐渐增强,并且在出壳初期的第2周内增加尤为显著,并且在21日龄时基本达到成熟水平。
To investigate the establishment of immune function in chicken respiratory-associated lymphoid tissue(RALT)and the immune state at embryonic stage and early stage after hatching,the mRNA expression levels of IFN-γand IL-2 in chicken were detected by real-time PCR.The results showed that the expression levels of cytokines IL-2 and IFN-γmRNA in RALT were extremely low in the embryonic stage and one day and four days post hatch.The expression levels of IL-2 and IFN-γmRNA gradually increased with age,and rapidly increased to higher levels at day 14,and reached the peak at day 21.The expression level of IFN-γin RALT was much higher than that of IL-2 in each period.In addition,the expression levels of IL-2 and IFN-γmRNA in the larynx,trachea and lungs were significantly higher than those in other organs during the same period.The results illustrated that the cellular immune function of RALT gradually enhanced with the growth and development,and the increase was especially significant in the second week after the initial exudation,and basically reached the maturity level at day 21.
