大豆品种安豆1498对大豆疫霉菌株PsJS2的抗性遗传分析及基因定位 被引量:1

摘要 为发掘新的抗病基因以增强大豆对疫霉根腐病的抗性,本研究以大豆品种安豆1498和Williams为材料,采用下胚轴创伤接种法鉴定二者对Race1、Race3、Race4、NKI、USAR2、Ps41-1、PsMC1和PsJS2等8个大豆疫霉菌株的抗性;对安豆1498与Williams杂交组合获得167个F2∶3代家系群体接种PsJS2菌株,进行抗性遗传分析;并利用618个SSR分子标记对抗性基因进行初步定位。结果表明:安豆1498对Race1、Race3、Race4、Ps41-1、PsMC1和PsJS2等6个大豆疫霉菌株表现为抗病,表明安豆1498是一个优良的疫霉根腐病抗性材料;抗性遗传分析结果显示安豆1498和F1代都表现抗病,Williams表现感病,F2∶3代群体中抗病:分离:感病株数表现为1∶2∶1的分离比,表明安豆1498对大豆疫霉菌株PsJS2的抗性由单个显性基因控制,暂命名为Rps1498;基因定位结果显示Rps1498与大豆3号染色体(N连锁群)上的5个SSR标记(Satt152、Satt631、Sat186、Satt683和Satt624)连锁,通过分子作图分析Rps1498位于SSR标记Sat186和Satt683之间,遗传距离分别为5.1和9.3 cM。 In order to discover new disease resistance genes to enhance soybean resistance to Phytophthora root rot(PRR).In this study, we used the hypocotyls inoculation technique to identify the resistance of soybean cultivars Andou 1498 and Williams to eight Phytophthora sojae(P.sojae) including Race1,Race3,Race4,NKI,USAR2,Ps41-1,PsMC1 and PsJS2.The F2∶3 generation population derived from the cross of Williams×Andou 1498 were inoculated P.sojae PsJS2 for each generation of the cross of Andou 1498 and Williams for resistance genetic analysis.618 SSR molecular markers were used for preliminary mapping of antagonistic genes.The results showed that Andou 1498 was resistant to six soybean Phytophthora strains, such as Race1,Race3,Race4,Ps41-1,Psmc1 and Psjs2,indicating that Andou 1498 was an excellent material resistant to Phytophthora root rot.The results of resistance genetic analysis showed that both the Andou 1498 and F1 generations were resistant to P.sojae PsJS2,Williams was susceptible, and the F2∶3 generation showed a segregation ratio of 1∶2∶1(resistance: segregation: susceptible).The resistance of Andou 1498 to P.sojae PsJS2 is controlled by a single dominant gene, temporarily named Rps1498.The gene mapping results showed that Rps1498 is linked to 5 SSR markers on soybean chromosome 3(N linkage group) including Satt152,Satt631,Sat186,Satt683 and Satt624.Basis of linkage analysis with SSR markers the Rps1498 was located on molecular linkage group N and flanked by Sat186 and Satt683 with genetic distances 5.1 and 9.3 cM,respectively.
