摘要
目的研究长链非编码RNA(lncRNA)溶质载体家族8成员A1反义RNA 1(SLC8A1-AS1)在脓毒症大鼠心肌组织中的表达及其对炎症因子分泌的影响。方法通过qRT-PCR方法检测脓毒症大鼠心肌组织中SLC8A1-AS1的表达。将心肌H9C2细胞分成对照组(Control)、脂多糖诱导组(LPS)、Vector+LPS(转染阴性对照载体,LPS诱导处理)、SLC8A1-AS1+LPS(转染SLC8A1-AS1过表达载体,LPS诱导处理)、SLC8A1-AS1+LPS+PMA组[转染SLC8A1-AS1过表达载体,核因子-κB(NF-κB)信号激活剂和LPS诱导处理]。酶联免疫吸附测定(ELISA)法检测细胞分泌的白细胞介素1β(IL-1β)、白细胞介素(IL-6)、肿瘤坏死因子-α(TNF-α)的水平。结果脓毒症大鼠心肌组织中SLC8A1-AS1表达降低(P<0.05)。与对照组比较,LPS组细胞分泌的IL-1β、IL-6、TNF-α增多;与Vector+LPS组比较,SLC8A1-AS1+LPS组细胞分泌的IL-1β、IL-6、TNF-α减少;与SLC8A1-AS1+LPS组比较,SLC8A1-AS1+LPS+PMA组细胞分泌的IL-1β、IL-6、TNF-α增多(P<0.05)。结论SLC8A1-AS1在脓毒症大鼠心肌组织中表达下降,上调SLC8A1-AS1可通过抑制NF-κB信号,减少心肌细胞凋亡和炎症因子分泌。
Objective To study the expression of long non-coding RNA(lncRNA)solute carrier family 8 member A1 antisense RNA1(SLC8A1-AS1)in myocardial tissue of sepsis rats and its effect on secretion of inflammatory factors.Methods qRT-PCR was used to detect the expression of SLC8A1-AS1 in myocardial tissue of sepsis rats.Cardiac H9C2 cells were divided into Control,LPS(LPS induction treatment),Vector+LPS(transfection negative control vector,LPS induction treatment),SLC8A1-AS1+LPS(transfection SLC8A1-AS1 overexpression vector,LPS induction treatment),SLC8A1-AS1+LPS+PMA group[transfected with SLC8A1-AS1 overexpression vector,nuclear factor kappa-B(NF-κB)signal activator and LPS induction treatment].Enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of IL-1β,IL-6 and TNF-αsecreted by cells.Results The expression of SLC8A1-AS1 in myocardial tissue of sepsis rats was decreased(P<0.05).Compared with the control group,the secretion of IL-1β,IL-6 and TNF-αwere increased in the LPS group;and compared with the Vector+LPS group,the secretion of IL-1β,IL-6 and TNF-αwere decreased in the SLC8A1-AS1+LPS group;and compared with the SLC8A1-AS1+LPS group,the secretion of IL-1β,IL-6 and TNF-αwere increased in the SLC8A1-AS1+LPS+PMA group(P<0.05).Conclusion The expression of SLC8A1-AS1 was decreased in myocardial tissue of sepsis rats.Up-regulation of SLC8A1-AS1 can secrete inflammatory factors by inhibiting NF-κB signaling.
