摘要
目的观察果王素(Kiwi fruit essence)对C57BL/6J小鼠接种Lewis肺腺癌细胞后移植瘤生长的影响并探讨其作用机制。方法将32只接种Lewis肺腺癌细胞的C57BL/6J小鼠按随机数字表法分成4组:对照组、低剂量果王素组(60mg/kg)、中剂量果王素组(120mg/kg)、高剂量果王素组(240mg/kg),每组8只。Lewis肺腺癌细胞接种于小鼠后第4天,对照组给予0.3ml生理盐水灌胃,每日1次,果王素组分别给予0.3ml相应剂量药物进行干预,每日1次,小鼠于接种肿瘤细胞后第24天均断颈法处死并剥离皮下移植瘤。采用实时荧光定量PCR(RT-PCR)检测小鼠移植瘤瞬时受体电位M8(Transient receptor potential melastatin 8,TRPM8)mRNA表达水平, Western blot法检测TRPM8蛋白表达水平。结果与对照组相比,果王素组小鼠皮下移植瘤体积、质量均不同程度减小,皮下移植瘤组织TRPM8 mRNA和蛋白表达均不同程度降低(P<0.05)。与低剂量果王素组相比,中剂量和高剂量果王素组小鼠皮下移植瘤体积、质量均减小,皮下移植瘤组织TRPM8 mRNA和蛋白表达均降低,且高剂量组各指标降低更明显(P<0.05)。结论果王素可通过抑制TRPM8表达抑制肺腺癌小鼠移植瘤的生长。
Objective To observe the effect of kiwi fruit essence on the growth of transplanted tumor in Lewis lung adenocarcinoma mice and investigate its mechanism. Methods 32 C57 BL/6 J mice inoculated with Lewis lung adenocarcinoma cells were randomly divided into 4 groups according to random number table method, control group, low-dose kiwi fruit essence group(60 mg/kg), medium-dose kiwi fruit essence group(120 mg/kg) and high-dose kiwi fruit essence group(240 mg/kg) with 8 mice in each group. On the 4 th day after Lewis lung adenocarcinoma cells were inoculated on the mice, the control group was given 0.3 ml normal saline by gavage once a day and the kiwi fruit essence group was given 0.3 ml corresponding dose of drugs for intervention once a day, respectively. The mice were sacrificed by neck breaking method and stripped the subcutaneous transplanted tumor on the 24 th day after tumor cells were inoculated. Real time fluorescence quantitative PCR was used to detect the expression level of transient receptor potential melastatin 8(TRPM8) mRNA and Western blot was used to detect the expression level of TRPM8 protein. Results Compared with the control group, the volume and mass of subcutaneous transplanted tumor in the kiwi fruit essence group decreased, and the mRNA and protein expression of TRPM8 in subcutaneous transplanted tumor decreased(P<0.05). Compared with the low-dose kiwi fruit essence group, the volume and mass of subcutaneous transplanted tumor in the medium-dose and high-dose kiwi fruit essence group decreased, the expression of TRPM8 mRNA and protein in subcutaneous transplanted tumor decreased, and the indexes in the high-dose kiwi fruit essence group decreased more significantly(P<0.05). Conclusion Kiwi fruit essence can inhibit the growth of transplanted tumor in lung adenocarcinoma mice by inhibiting the expression of TRPM8.
