摘要
试验研究枯草芽孢杆菌(Bacillus subtilis)产纤维素酶(CMCA)的活性条件。以杨树叶、酒糟、红薯秧、麸皮为主要试验碳源,通过单因素试验确定最佳碳源,并通过单因素试验和响应面法优化分析碳源添加量、产酶温度、装液量对纤维素酶活力的影响。结果表明,产酶最佳的培养条件为装液量100.510 m L、产酶温度36.995℃、碳源添加量为7.049 g/L。在此条件下,酶活力最高,为3.662 U/mL。
The activity conditions of cellulase(CMCA)produced by Bacillus subtilis were studied.Distillers’grains,sweet potato seedlings,poplar leaves and bran were used as the main test carbon sources.The best carbon source was determined by single factor test.The effect of the amount of carbon source,temperature and liquid volume on cellulase activity were optimized by single factor test and response surface method.The results showed that the optimal culture conditions were liquid volume 100.510 mL,temperature 36.995℃,carbon source 7.049 g/L.Under the optimal conditions,the enzyme activity was up to 3.662 U/mL.
