基于UPLC指纹图谱和多成分定量的茵陈配方颗粒质量控制研究 被引量:6

Quality Control of Artemisiae Scopariae Herba Dispensing Granules Based on UPLC Fingerprints and Multi-component Content Determination

摘要 目的建立茵陈配方颗粒超高效液相色谱(UPLC)指纹图谱及多成分含量测定方法,为其质量评价提供依据。方法采用Acclaim^(TM) RSLC 120 C18色谱柱(2.1 mm×100 mm,2.2μm),以乙腈-0.05%磷酸水溶液为流动相,梯度洗脱,流速0.4 mL/min,检测波长327 nm,柱温30℃,进样量1μL,建立14批茵陈配方颗粒指纹图谱,并对指标成分进行含量测定。结果建立了茵陈配方颗粒的UPLC指纹图谱,确定了9个共有峰,指认出其中8个成分,分别为新绿原酸、绿原酸、隐绿原酸、咖啡酸、1,3-二咖啡酰奎宁酸、3,4-二咖啡酰奎宁酸、3,5-二咖啡酰奎宁酸和4,5-二咖啡酰奎宁酸,并进行了含量测定,各成分色谱峰分离较好,线性关系、重复性、稳定性、回收率均符合要求。结论本研究建立的UPLC指纹图谱及多成分含量测定方法简便、准确,重复性良好,可为茵陈配方颗粒的质量评价提供依据。 Objective To establish the UPLC fingerprints of Artemisiae Scopariae Herba dispensing granules and its multi-component content determination method;To provide a basis for its quality evaluation.Methods UPLC separation was performed to establish the fingerprints of 14 batches of Artemisiae Scopariae Herba dispensing granules on an Acclaim^(TM) RSLC 120 C18 column(2.1 mm×100 mm,2.2μm)with acetonitrile-0.05% phosphoric acid aqueous solution as the mobile phase,gradient elution.The flow rate was 0.4 mL/min;the detective wavelength was 327 nm;the column temperature was 30℃;the injection volume was 1μL.The index components were determined.Results The UPLC fingerprints of Artemisiae Scopariae Herba dispensing granules were established.There were 9 common peaks as the characteristic peaks in HPLC,eight of which were identified as gneochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,caffeic acid,1,3-dicaffeoylquinic acid,3,4-dicaffeoylquinic acid,3,5-dicaffeoylquinic acid and 4,5-dicaffeoylquinic acid.The chromatographic peaks of each component were well separated.The linear relationship,repeatability,stability,and recovery rate all met the requirements.Conclusion The UPLC fingerprints and multi-component content determination method is simple,accurate,and repeatable,which can provide a basis for quality evaluation of Artemisiae Scopariae Herba dispensing granules.
