摘要
目的研究水栀子Gardenia jasminoides var.radicans的萜类成分及其肾细胞保护活性。方法采用硅胶、ODS等柱色谱和制备液相色谱等手段分离纯化,根据波谱数据鉴定化合物结构。采用MTT法探究所得化合物对脂多糖(LPS)诱导的大鼠肾小管上皮(NRK52e)细胞损伤的干预作用。结果分离并鉴定出17个化合物,分别为3-oxo-2,6,6-trimethylcyclohex-l-ene-lcarboxylic acid(1)、rehmapicrogenin monomethyl ester(2)、jasminodiol(3)、4-hydroxy-2,6,6-trimethylcyclohex-1-enecarboxylic acid(4)、jasminoside A(5)、jasminoside E(6)、6'-O-trans-sinapoyljasminoside L(7)、6'-O-trans-sinapoyljasminoside B(8)、6'-O-(3-methoxyl-4-hydroxyl-coumaroyl)-epijasminoside B(9)、jasminoside S(10)、11-氧代熊果酸(11)、钩藤酸E(12)、(1S*,17R*,20R*,24Z)-1,21,26-trihydroxycycloart-24(25)-en-3-olide(13)、3β,6β,23-三羟基齐墩果酸(14)、冬青酸(15)、栀子花甲酸(16)、栀子花乙酸(17)。结论化合物1为新天然产物,化合物2、9、11为首次从栀子属中分离得到,其余化合物均为首次从该植物中分离得到。化合物1、2、9、14对LPS诱导的NRK52e细胞损伤具有明显的保护作用。
OBJECTIVE To study the terpenoids from Gardenia jasminoides var.radicans and their renoprotective activity.METHODS The compounds were isolated and purified by silica gel and ODS column chromatography,as well as pre-HPLC.Their structures were identified by spectral analysis.The effects of isolated compounds on NRK52e cells injury induced by LPS were investigated by MTT method.RESULTS Seventeen compounds were isolated and their structures were identified as 3-oxo-2,6,6-trimethylcyclohex-l-ene-l-carboxylic acid(1),rehmapicrogenin monomethyl ester(2),jasminodiol(3),4-hydroxy-2,6,6-trimethylcyclohex-1-ene-1-carboxylic acid(4),jasminoside A(5),jasminoside E(6),6'-O-trans-sinapoyljasminoside L(7),6'-O-trans-sinapoyljasminoside B(8),6'-O-(3-methoxyl-4-hydroxyl-coumaroyl)-epijasminoside B(9),jasminoside S(10),11-oxo-ursolic acid(11),uncarinic acid E(12),(1S*,17R*,20R*,24Z)-1,21,26-trihydroxycycloart-24(25)-en-3-olide(13),3β,6β,23-trihydroxyolean-12-en-28-oic acid(14),rotundic acid(15),gardenolic acid A(16),and gardenolic acid B(17).Compounds 1,2,9,and 14 could significantly alleviate the injury of NRK52E cells induced by LPS.CONCLUSION Compound 1 is a new natural product,compounds 2,9,and 11 are isolated from genus Gardenia for the first time,and all the other compounds are isolated from this plant for the first time.Compounds 1,2,9,and 14 exhibit protective effects against LPS-induced damage to NRK52e cells.
