hsa-miR-26b靶向ERα调控人卵巢癌HO-8910细胞周期的作用机制

摘要 目的探讨hsa-miR-26b靶向ERα基因调控人卵巢癌HO-8910细胞周期的分子机制.方法HO-8910细胞分为3组:空白对照组(control组)、miR-26b激动剂组(miR-26b mimic组)和miR-26b激动剂阴性对照组(mimic NC组),Real-time PCR检测miR-26b转染效率,细胞增殖试验(CCK8)检测细胞活性,流式细胞术检测细胞周期,Real-time PCR和Western Blot检测ERα mRNA和蛋白的表达水平,双荧光素酶报告基因检测miR-26b与ERα 3′UTR的相互作用.结果CCK8显示miR-26b可抑制HO-8910细胞增殖活性,流式细胞术显示细胞增殖活性抑制与G1期阻滞相关,Real-time PCR和Western Blot显示其可抑制ERα mRNA和蛋白表达,并抑制Cyclin D1和CDK6蛋白表达水平,双荧光素酶报告基因检测显示miR-26b与ERα 3′UTR可相互结合.结论miR-26b通过负调控ERα表达水平,下调Cyclin D1和CDK6以阻滞HO-8910细胞G1期,从而抑制细胞的增殖活性.
